      Thin-layer chromatography — an image processing method for the determination of acidic catecholamine metabolites

HPTLC of the acidic catecholamine metabolites (1) 3,4-dihydroxymandelic acid, (2) epinephrine, (3) vanillylmandelic acid, (4) 3,4-dihydroxyphenylacetic acid and (5) homovanillic acid in human urine on RP-18 with citrate buffer (pH 3.0) - methanol - formic acid 48:2:5. Detection by spraying with 0.02 % 2,2-diphenyl-1-picrylhydrazyl radical solution in ethanol. Quantification by digital imaging processing. The hRF values of (1) to (5) were 84, 65, 59, 47 and 22, respectively. Linearity was in the range of 30-150 ng/zone for (1), (2) and (4), 90-450 ng/zone for (3) and 60-300 ng/zone for (5). The intermediate intra-day and inter-day precisions were below 3 % (n=3) for (1) to (5). The LOD and LOQ were 33 and 36 ng/zone for (1), 36 and 42 ng/zone for (2), 103 and 120 ng/zone for (3), 13 an 17 ng/zone for (4) and 45 and 54 ng/zone for (5), respectively. Recoveries for the compounds were between 94.6 and 105.7 %.

TLC of chlorpromazine hydrochloride, trifluoperazine dihydrochloride, promazine hydrochloride, and doxepin hydrochloride on silica gel with 1-butanol - aqueous ammonia solution 5:1 or cyclohexane - acetone - diethylamine 8:1:1 after saturation with mobile phase vapor. Detection by inspection under UV light at 254 nm. Scanning densitometry was performed at 254 nm and at the wavelengths of maximum absorbance of the substances.

The method has a range from 100 - 300 ng/zone. The limit of detection and quantification was 25 ng and 100 ng/zone, respectively. The method was applied for analysis of metformin hydrochloride in multi-component formulations containing glipizide, gliclazide and glibenclamide as well as in serum.

      SIMULTANEOUS HPTLC ESTIMATION OF AMBROXOL HCl AND CETIRIZINE HCl IN THEIR COMBINED DOSE TABLET

      Asian J. Chem. 19(2), 1487-1493 (2007). TLC of ambroxol HCl and cetirizine HCl on silica gel with methanol - ethyl acetate - toluene - ammonia 3:10:12:1. Quantitative determination by absorbance measurement at 231 nm. The hRf value of ambroxol HCl was 78 and of cetirizine HCl 40. The calibration curve response was observed between 4 and 10 µg for ambroxol HCl and 0.4 and 0.8 µg for cetirizine HCl via peak height and area. The percent drug estimated for ambroxol HCl and cetirizine HCl from marketed formulation was 99.9, 100.2 by height and 100.8, 99.1 by area respectively. Recovery of ambroxol HCl via peak height and via peak area was 102.7 and 100.0 % respectively, and for cetirizine HCl 99.3 and 98.1 %.
