      A High Performance Thin Layer Chromatography (HPTLC) Method for the estimation of Gallic Acid from Emblica officinalis (AMLAKI)

      59th Indian Pharmaceutical Congress C-220, 277, (2007). HPTLC of gallic acid in fruits of Emblica officinalis, on silica gel with touene - ethyl acetate - acetic acid - formic acid 4:9:4:3. Densitometry at 277 nm for quantitative evaluation. The method was linear in the range of 10 and 1000 ng/mL, with an average recovery of 101.4 %. The method was found suitable for routine quality check-up of Emblica officinalis.

      TLC determination of strychnine and brucine of Strychnos nux vomica in Ayurveda and homeopathy drugs

Quantitative estimation of the two major alkaloids strychnine and brucine in Strychnos nux vomica by TLC on silica gel with chloroform - ethyl acetate - diethyl amine 1:17:2. The hRf values for strychnine and brucine were 55 and 42 and selectivity regarding matrix was given. Recovery was between 93.1 and 99.8 % for strychnine and between 96.9 and 99.5 % for brucine. The limit of detection and quantification for strychnine was 2 and 8 ng, respectively, and for brucine 2 and 9 ng, respectively.

      A reverse phase HPLC-UV and HPTLC methods for determination of plumbagin in Plumbago indica and Plumbago zeylanica

      Ind. J. of Pharma Sci. 70(6), 844-847 (2008). HPTLC of plumbagin in alcoholic root extracts of Plumbago indica and Plumbago zeylanica on silica gel with n-hexane - ethyl acetate 4:1. The hRf value of plumbagin was 67. Quantitative determination by absorbance measurement at 265 nm. Plumbago indica showed significantly higher plumbagin contents than P. zeylanica. The HPTLC method was the method of choice for simultaneous analysis of several samples, whereas the RP-HPLC method was very sensitive.

Densitometric evaluation at 285 nm. The method was linear in the range of 100-500 ng/band for both compounds. The average recovery was 99.9-102.1 for both compounds and there was no interference with additives present in tablet dosage forms.

For quality control, TLC on silica gel 1) for borneol, with cyclohexane – ethyl acetate 33:7, detection by spraying with 1 % vanillin in sulfuric acid – ethanol 1:4, heating at 105 °C until the zones are visible in daylight; 2) for Cortex Phellodendri Chinensis and the standard berberine hydrochloride, with benzene – ethyl acetate – methanol – isopropanol – concentrated ammonia 12:6:3:3:1, detection under UV 366 nm; 3) for Rheum palmatum L., Reynoutria japonica Houtt, and the standard emodin, with petroleum ether (30-60 °C) – ethyl formate – formic acid 15:5:1, detection under UV 366 nm before and after exposure to ammonia vapors.
