      Thin-layer chromatography and electrospray mass spectrometry coupled using a surface sampling probe

      Anal. Chem. 74, 6216-6223 (2002). A combined surface sampling probe/electrospray emitter was used for the direct readout of TLC plates by electrospray MS. HPTLC of methylene blue, crystal violet, and rhodamine 6G on RP-18 with methanol – tetrahydrofuran 32 containing 50-100 mM ammonium acetate, followed by MS detection in positive ion mode. HPTLC of fluorescein, naphtholblue black, and fast green FCF on PR-18 with methanol – water 73, followed by MS detection in negative ion mode. Drying by heating at 110°C for 30 min in an oven. Acquisition of mass spectra of components of individual bands was shown by manual stepping to and sampling from specific locations within the bands. Computer-controlled scanning of lanes was illustrated by using multiple ion monitoring in positive and negative ion modes. Readout resolution, the limits of scan speed, detection levels, TLC phase (restricted to nonpolar phases), and eluting solvents were investigated.

      Dreidimensionale Darstellung von Dünnschicht-Chromatogrammen. (Three-dimensional graphic presentation of thin-layer chromatograms). Apart from quantification storage of chromatograms is performed graphically in many cases, this should be done in a three dimensional way. For UV-detection, acquiring and presentation of data is described. Example 1: two-dimensional HPTLC of amitriptyline derivatives on silica with 1) chloroform - methanol - 25 % NH3 90:15:1.2) toluene; example 2: TLC of structurally similar components on silica with chloroform - methanol 9:1; example 3: TLC on silica with chloroform - methanol diethylamine 90:10:1.

Analysis of the antioxidants ascorbic acid (1), gallic acid (2), caffeic acid (3), quercetin (4), and biogenic amines (–)-epinephrine (5), (–)-norepinephrine (6), isoprenaline hydrochloride (7), dopamine hydrochloride (8), 3,4-dihydroxy-d-phenyl-alanine (d-dopa) (9), and 3′,4′-dihydroxy-2-(methylamino)acetophenone hydrochloride (adrenalone) (10) applied as individual zones in different concentrations on silica gel, without development. Radical-scavenging activity was measured by spraying with 0.03 % 2,2-diphenyl-1-picrylhydrazyl (DPPH radical reagent) solution in methanol and stored in dark at 22 °C for 5 min, followed by scanning in a dark place. Image data was converted into chromatograms, from which the spot area as a function of the reaction time was monitored. The title "Chromatographic approach" is misleading, as there is no chromatography.

Quantitation of tanshinone ll A by HPLC.
