      A pharmacognostic study on Sphaeranthus indicus

HPTLC of the ethyl acetate soluble fraction of aqueous alcoholic Sphaeranthus indicus extracts, on silica gel with n-hexane - diethyl ether 3:7. The plant extract and the marker (7-OH-eudesmanolide) were chromatographed simultaneously. Quantitative determination by absorbance measurement at 213 nm.

Detection by dipping into vanillin - sulfuric acid reagent, followed by heating at 90 ºC for 3 min. The hRf values for (1) and (2) were 56 and 45. Bioautography by dipping into S. aureus suspension for 10 s. After drying for 2 min followed by incubation at 37 ºC for 17 h the plates were dipped in an aqueous solution of MTT (3-[4,5-dimethylthiazol-2-yl]-2,5-diphenyltetrazolium bromide 50 mg/80 mL) for 10 s and incubated at 37°C for 2 h. Inhibition zones appeared as pale yellowish zones against blue background.

      High-performance thin-layer chromatography-direct bioautography as a method of choice for alpha-amylase and antioxidant activity evaluation in marine algae

Evaluation of the phenolic and stigmasterol content and comparison of the antioxidant and antidiabetic activities by HPTLC combined with DPPH* free radical method and α-amylase assay towards ethanol and ethyl acetate extracts of 10 marine macroalgae species (3 chlorophyta, 4 phaeophyta and 3 rhodophyta). HPTLC on silica gel with n-hexane – ethyl acetate – acetic acid 20:9:1 over 80 mm. Detection either by dipping into 0.4 % DPPH* solution followed by storing in the dark for 30 min before evaluation, or by dipping into anisaldehyde – sulfuric acid reagent or neutralized ferric chloride solution (prepared by adding dilute sodium hydroxide to freshly prepared 2 % ferric chloride in methanol until precipitation occurs, followed by filtering), both followed by heating at 100 °C for 10 min. Quantitative evaluation by absorbance measurement at 470 nm for detection of α-amylase inhibition and at 550 nm for stigmasterol. The results showed higher antioxidant activity in the ethyl acetate extracts than in ethanol extracts, and higher amounts of fucoxanthin, stigmasterol and α-amylase inhibitory activities in ethyl acetate extracts. The results proved the relation of higher α-amylase inhibition in ethyl acetate extracts due to the presence of triterpenes and sterols, and no contribution of fucoxanthin to it.

      Virgaureasaponin 1, a new bisdesmosidic triterpenoid saponin of Solidago virgaurea L. Pharmazie 42, 541-543 (1987). TLC of saponins on silica with chloroform - methanol - water 15:10:2, with chloroform - methanol 9:1 and with n-butanol - ethanol - water 4:1:5 (upper phase). Detection reagents: blood gelatin for saponins, anisaldehyde - sulfuric acid for saponins and sapogenins and thymol - sulfuric acid for sugars. A new triterpenesaponin isolated and identified.
