HPTLC of quercetin and kaempferol in horizontal chambers on silica gel (prewashed with methanol) with four mobile phases, e.g.1,4-dioxane - toluene - 85 % acetic acid 6:24:1 or on cellulose with five mobile phases. Evaluation under UV light at 254 and 366 nm before and after spraying with a 2 % solution of zirconium (IV) dichloride oxide in methanol. Quantitative determination by absorbance measurement at 373 for quercetin and at 347 nm for kaempferol.

      Development and validation of HPTLC method for estimation of tacrolimus in formulations

      Drug Development and Industrial Pharmacy 35, 440-448 (2009). TLC of tacrolimus on silica gel with toluene - acetonitrile - glacial acetic acid 60:40:1. For quantitative evaluation the plate was derivatized with anisaldehyde sulfuric acid reagent and scanned at 675 nm. Using this method a compact spot was obtained at an hRf value of 40. The linearity range was 100-800 ng/band. The method is suitable for routine analysis of formulations.

      Development and validation of a novel HPTLC method for simultaneous estimation of beta-sitosterol-D-glucoside and withaferin-A

TLC of beta-sitosterol-D-glucoside and withaferin-A in Withania somifera formulations on silica gel with chloroform - methanol 4:1. The hRf value of beta-sitosterol-D-glucoside was 21 and of withaferin-A 59. Quantitative absorbance measurement at 207 nm. The method was linear in the range of 50-500ng/band for beta-sitosterol-D-glucoside and in the range of 5-50 ng/band for withaferin-A.

HPTLC of eight natural products viz. stigmasterol (1), oleanolic acid-3-acetate (2), oleanolic acid (3), biochanin A (4), genistein (5), pratensein (6), chrysoeriol (7), and luteolin (8) in Cicer microphyllum on silica gel with n-hexane - ethyl acetate - formic acid 90:65:8. Detection by dipping into a solution of cerric ammonium sulfate reagent, followed by heating at 105 °C for 5 min. Quantitative determination by absorbance measurement at 366 nm. The hRF values for compounds (1) to (8) were 98, 89, 78, 70, 63, 42, 33 and 4, respectively. Linearity was in the range of 100-800 ng/zone for (1) to (8). LOD and LOQ were 60 and 90 ng/zone for (1), (5) and (8), 50 and 90 ng/zone for (2), 90 and 110 ng/zone for (3), 70 and 100 ng/zone for (4) and (6) and 80 and 110 ng/zone for (7). The intermediate precision was below 1.5 % (n=3). Recovery was in the range of 98-100 %.
