      The effect of metal salts on the separation of carbohydrates by TLC

TLC of carbohydrates on silica gel plates impregnated by dipping in methanolic solutions or by use of mobile phase additives; the mobile phases were mixtures of acetonitrile, 2-propanol, or acetone with water or aqueous solutions of the metal salt. Plates were developed in horizontal chambers. Post-chromatographic derivatization was performed by spraying with a solution of diphenylamine (2 g), aniline (2 mL), 85% phosphoric acid (10 mL), and methanol (100 mL) and drying for 5 min at 105°C.

Detection by spraying with alpha-naphthol–sulfuric acid sugar-detection reagent followed by heating at 110 ºC for 5 min. Densitometry of glucose and raffinose at 515 nm. HPTLC of amino acids with four different TLC systems: on silica gel with preadsorbent zone, on RP-18 with concentrating zone, on cellulose F with n-butanol – acetic acid – water 3:1:1, and on ion-exchange sheets with citrate buffer of pH 3.3 (84 g citric acid monohydrate, 16 g NaOH, and 5.9 mL HCl 37% per L). Detection by spraying with ninhydrin reagent, followed by drying in air for 30 min and heating for 10 min at 110 ºC. Quantification by densitometry at 495 nm for histidine and 610 nm for tryptophan.

Detection of (1) on silica gel with a 1:1 mixture of 10 % α-naphthol in ethanol and 10 % sulfuric acid in water, followed by heating at 120 °C for 5-10 min; and on RP18 W by spraying with a 25 % methanolic solution of sulfuric acid, followed by heating at 120 °C for 5-10 min. Detection of (2) by spraying with 20 % aqueous solution of perchloric acid, followed by heating at 120 °C for 5 min. A comprehensive retention data set was obtained for fast selection of mobile phase compositions.

      High performance thin layer chromatography determination of cellobiosan and levoglucosan in bio-oil obtained by fast pyrolysis of sawdust

HPTLC of sugars in bio-oil fractions on silica gel with acetonitrile - water 4:1, or mixtures of butanol and formic acid, followed by detection with the aniline - diphenylamine - o-phosphoric acid reagent. The method allowed for the separation of the anhydrosugars levoglucosan and cellobiosan, as well as glucose, arabinose, xylose and cellobiose without the need of pre-treatment and pre-derivatization of samples. Volatile compounds present in bio-oil did not interfere with sugar analysis, and the detrimental effect of the complex bio-oil matrix on columns and detector lifetime is avoided by using disposable HPTLC plates. It was found that the concentrations of levoglucosan and cellobiosan in bio-oil samples obtained from Pinus radiata sawdust were ranged between 1.3-2.3 % and 1.0-2.0 % respectively, while a higher levoglucosan concentration was in a bio-oil sample obtained from native wood.

      (Purity test of sorbose and sorbitol with thin-layer chromatography on Silufol ready made plates.) Pharmazie 39, 689-690 (1984). Sorbitol and sorbose are separated from concomitant substances such as glucose and/or fructose, disaccharides and trisaccharides. The silica layer pretreated with 2O mM solution of sodium pyrosulfate, then developed with 1-propanol - ethyl acetate - water 27:3.5:1 and visualized after drying at 100 °C by spraying with lead tetraacetate.
