Determination of relationships between RF values and mobile phase composition for twenty moderately polar pesticides in systems of the type normal- and reversed phase HPLC and TLC.       Use of database of plots of pesticide retention (Rf) against mobile-phase compositions for fractionation of a mixture of pesticides by micropreparative Thin-Layer Chromatography

Relationships between Rf values and mobile phase composition have been determined for urea herbicides and fungicides in normal-phase systems (NP) of the type silica-nonpolar or weakly polar diluent (heptane, toluene, diisopropyl ether) – polar modifier (ethyl acetate, tetrahydrofuran, dioxane, ethyl-methyl ketone and 2-propanol). These relationships constitute a retention database which has enabled to choose optimum systems for preliminary fractionation of a multicomponent mixture of pesticides by zonal micropreparative TLC. The mixture was applied from the edge of the layer in the “frontal + elution” mode which increased the separation efficiency because of displacement effects. The separated simpler fractions were applied to a silica plate and rechromatographed. The plate was videoscanned, furnishing a real picture of the plate showing preliminary separation of the simpler pesticide fractions. Complete separation of the fractions was carried out by two-dimensional thin-layer chromatography on plates with chemically bonded-cyanopropyl silica stationary phase using non-aqueous eluent in the first direction and aqueous reversed-phase eluent in the second direction.

      A new spray reagent for selective detection and quantification of dichlorvos in bluish tinged maize grains by TLC-spectrophotometry

      J. Planar Chromatogr. 24, 113-115 (2011). TLC of dichlorvos on silica gel with cyclohexane - acetone - methanol 16:6:1. Detection by spraying with 2 % sodium hydroxide solution and subsequent spraying with 2 % thiobarbituric solution followed by heating at 90 °C for 10 min. The hRf of dichlorvos was 50. The limit of detection was approximately 18 µg/zone. Spectrophometric analysis of dichlorvos was performed by measuring the absorbance of the sample solution. The linearity was in the range of 50-350 µg/mL dichlorvos. On alkaline hydrolysis dichlorvos forms dimethylphosphoric acid and dichloroacetaldehyde; the latter reacts with 2-thiobarbituric acid to give a sharp pink spot. The reagent is selective for dichlorvos, and does not react with other organophosphorus, organochlorine, carbamate, and synthetic pyrethroid insecticides.

      Determination of organophosphorus pesticides in rice by TLC

      Asian Journal of Chemistry 23(5), 2011-2013 (2011). Methods are reported for determination of organophosphorus pesticides using TLC-cholinesterase inhibition as well as GC-FID. The powdered rice sample was initially extracted with ethyl acetate. The extract was cleaned up by GPC, eluted with cyclohexane - ethyl acetate 1:1, evaporated, and taken up in acetone. This extract was analysed by TLC on silica gel with ethyl acetate to a developing distance of 10-12 cm. After development the plate was air dried, exposed to bromine vapors, sprayed with enzyme solution and incubated at 37 °C for 30 min and sprayed with the reagent. White spots appeared against bluish-red background. The sample was also analysed by GC and the GC method was very sensitive. However, the TLC method is recommended for preliminary screening of samples.

      Influence of substrate on the F-2 and T-2 production of fusarium species

      Acta Alimentaria 16, 29-43 (1967). Twenty-one identified strains of fusarium sp. were examined for their ability to produce F-2 and T-2 toxin on sterilized, moistened rice and corn grains. The F-2 and T-2 toxin production was estimated by TLC and confirmed by GLC. TLC of F-2 toxin on silica with toluene - ethyl acetate - chloroform 95:55:50 and chloroform - acetone 97:3; after development plates heated for 5 min. at 120 °C, detection of zearalenone by UV 366 and 254 nm. Detection of F-2 toxin by spraying with 1 % 4-methoxy - benzene - diazonium- fluoroborate and heating for 10 min at 110 °C. Sensitivity: 0.03 mg/spot. TLC of T-2 toxin on silica with toluene - ethyl acetate - 90 % formic acid 6:3:1; spraying with 20 % sulfuric acid in 96 % ethanol and heating for 10 min at 110 °C, detection by UV at 366 nm. Sensitivity: 0.05 mg/spot.
