      Imaging of thin-layer chromatograms using matrix-assisted laser desorption/ionization mass spectrometry

      Anal. Chem. 67, 4565-4570 (1995). Introduction of a new approach for direct coupling TLC with matrix-assisted laser desorption/ionization mass spectrometry. To avoid excessive analyte spreading along a TLC plate, a matrix layer was first generated on a different substrate, from which it was transferred to the plate. The transfer was accomplished by pressing the matrix onto a previously wetted TLC plate. Successful direct imaging of silica gel TLC plates with high spatial resolution (250-500 µm) and high sensitivity has been demonstrated for several compounds, including rhodamine B, guinea green B, bradykinin, and angiotensin II, in the nanogram range. The detection limit was in the picogram range.

Report of the investigation and optimization of TLC/matrix-assisted laser desorption ionization (MALDI) coupling protocol, stressing on the fundamental coupling parameters which influence sensitivity and lateral analyte spreading, like extraction solvent selection, extraction time, and pressures. The upper analyte recovery limit, using extraction solvents with Rf values between 0.4 to 0.6 is estimated at 22%.

      A planar chromatographic method for preconcentration of impurities of low volatility by thermal desorption of the volatile main component

Description of a preconcentration method based on continuous elution and intense thermal desorption of the main volatile component in a narrow zone on the plate. A fiftyfold concentration of a model dye mixture from an aqueous solution was achieved on a TLC plate in 20 - 30 min. Silica gel, propan-2-ol as mobile phase and quantitation by densitometry at 530 nm. The technique enables determination of the amounts of common metals in drinking water.

      Use of multiple readings to increase the sensitivity of phosphor image detection in TLC

In thin-layer radiochromatography the high sensitivity of the phosphor imaging analyzer can be further increased by multiple reading of the image plates, because the latent image is not lost quantitatively in the reading process. Summing of the chromatograms obtained in successive readings results in increased signal-to-noise ratio. Thus, by use of this approach either the exposure time can be shortened or higher sensitivity can be achieved. TLC of a14C-labeled test substance in rat serum on silica gel with chloroform - n-hexane - ethanol - ammonia 75:15:9:1. Detection by radioluminography.
