Normal- and reversed-phase thin-layer chromatography of seven oral antidiabetic agents
      A. GUMIENICZEK*, H. HOPKALA, A. BERECKA, D. KOWALCZUK, (*Dept. of Med. Chem., Med. Univ. of Lublin, Chodzki Str. 6, 20-093 Lublin, Poland)

      J. Planar Chromatogr. 16, 271-275 (2003). TLC of chlorpropamide, tolbutamide, glibenclamide, metformin, pioglitazone, rosiglitazone, and repaglinide on silica gel and oxide with mixtures of chloroform, ether, and ethyl acetate. For more effective resolution aqueous ammonia or acetic acid was added to the mobile phase. Silica enabled better separation than alumina. Reversed-phase chromatography was performed on RP-18 with mixtures of acetonitrile or 2-propanol with phosphate buffer. For the separation of these drugs RP chromatography was more effective than use of normal-phase mode. Determination under UV 254 nm. Systematic optimization of separation conditions.

      P. G. SUNITHA, V. VAIDHYALINGAM, A. SHANTA, R. P. EZHIL MUTHU (Department of Pharmaceutical Chemistry, Madras Medical College, Chennai 600003, India)

Indian Drugs 41 (3), 149-152 (2004). HPTLC on silica gel with toluene - ethyl acetate 1:24. Quantitative determination by absorbance measurement at 260 nm. The linearity range is 100-500 µg/µL and recovery is 100.7 %. The method was validated and compared with HPLC. The results were comparable. Advantages of HPTLC in terms of handling many samples. Thus, a simple, fast and precise HPTLC method was developed for quantification of ebastine in tablets.

      H. HOPKALA, A. POMYKALSKI* (*Department of Medicinal Chemistry, Faculty of Pharmacy, Prof. Skubiszewski Medical Universit y of Lublin, 6 Chodzki St., 20-093 Lublin, Poland)

      J. Planar Chromatogr. 17, 383-387 (2004). TLC of fenbufen, ibuprofen, ketoprofen, diclofenac sodium, mefenamic acid, and tiaprofenic acid on silica gel by ascending and horizontal techniques, and on RP-18 in horizontal chambers. Good separation was achieved on silica gel by horizontal development with chloroform - methanol - 25 % ammonia 67:25:8; reversed phase chromatography on RP-18 with 0.15 mol/L phosphate buffer, pH 5.73 - 10 % CTMA-Br (N-cetyl-N,N,N-trimethylammonium bromide) in methanol 7:13 enabled better separation of the six drugs. Detection under UV light at 254 nm - for ibuprofen detection was best achieved after normal phase chromatography with 20 % aqueous sodium carbonate solution. A simple videodensitometric procedure was developed and validated. RSD for quantitation of fenbufen was 2.44 - 3.10 %.
