Huaiyu Wan pill is a herbal TCM for treating bleeding of rectum polyp, anal fissure, hemorrhage due to ulcerative colitis etc. For quality control, TLC on silica gel 1) for Gardenia jasminoides Ellis and the standard gardenoside, with ethyl acetate – acetone – formic acid – water 5:5:1:1, detection by spraying with 10 % sulfuric acid in ethanol and heating at 110 °C until the zones are visible in daylight; 2) for flowers and buds of Sophora japonica L. and the standard rutin, with ethyl acetate – formic acid – water 8:1:1, detection by spraying with 1 % aluminum trichloride in ethanol, detection under UV 366 nm; 3) for Paeonia suffruticosa Andr. and the standard paeonol, with cyclohexane - ethyl acetate 3:1, detection by spraying with 2 % ferric chloride in ethanol and heating until the zones are visible in daylight. Quantification of rutin by HPLC.

For quality control, TLC on silica gel 1) for Lobed Kudzuvine root and Pueraria lobata, with the upper phase of toluene – ethyl acetate – formic acid 5:3:1, detection under UV 366 nm; 2) for Hovenia acerba Lindl., with the upper phase of n-hexane – ethyl acetate – ethanol – ammonia 14:8:2:1, detection under UV 366 nm. Quantification of chlorogenic acid by HPLC.

      Simultaneous quantitation of piperine and piperlongumine in the fruit of Piper longum Linn

      by validated high-performance thin-layer chromatography-densitometric method. J. Planar Chromatogr. 27, 362-366 (2014). HPTLC of (1) piperine and (2) piperlongumine in the fruits of Piper longum on silica gel with n-hexane – ethyl acetate 4:1. Quantitative determination by absorbance measurement at 290 nm. The hRF values of (1) and (2) were 24 and 54, respectively. Linearities were between 100 and 500 ng/zone for (1) and between 200 and 1000 ng/zone for (2). The intermediate precisions were below 1 % (n=6) for both, (1) and (2). The LOD and LOQ were 40 and 80 ng/zone for (1) and 90 and 200 ng/zone for (2), respectively. Average recoveries for (1) and (2) were 99.2 and 99.3 %, respectively.

HPTLC identification of C. aethusifolia, C. intricate, C. brevicaudata, C. hexapeta, C. macropetala and C. mandshurica on silica gel with (A) petroleum ether (60-90°C) – ethyl acetate – acetone 9:3:1 at 25 °C and relative humidity of 50 %, detection by spraying with sulfuric acid – water 1:3, followed by heating at 100 °C until the spots were visible, or (B) chloroform – methanol – water 30:6:1 at 25 °C and relative humidity of 50 %, detection by spraying with sulfuric acid – water 1:3, followed by heating to 100 °C and viewing at UV 366 nm.

Quantitative determination of paeoniflorin, gardenoside, emodin, and chrysophanol by HPLC.

Quantitative determination by absorbance measurement at 242 nm. The hRF value for cucurbitacin was 67. Linearity was in the range of 200-1200 ng/zone. LOD and LOQ were 40 and 123 ng/zone, respectively. The intermediate precision was below 6 % (n=6). Recovery ranged between 95 and 96 %.

Detection by dipping into a solution of p-dimethyl aminobenzaldehyde in methanolic hydrochloric acid for 2 s, followed by heating at 110 °C for 3 min. Quantitative determination by absorbance measurement at 515 nm. The hRF value for protodioscin was 31. Linearity was in the range of 400-1400 ng/zone. LOD and LOQ were 100 and 400 ng/zone. The intermediate precision was below 5.1 % (n=3). Average recovery was 101 %.
