      Simultaneous quantification of flavonoids in Ginkgo biloba using RP-HPTLC densitometry method

HPTLC of flavonoids (quercetin and kaempferol) and biflavonoids (sciadopitysin, ginkgetin, and bilobetin) in aqueous methanolic extracts of Ginkgo biloba on RP-18 by double development with 1) acetonitrile - water - methanol - formic acid 20:20:1:0.005 and 2) acetonitrile - water - methanol - formic acid 20:17:1:0.005. Quantitative determination by densitometry in absorption mode at 254 nm. LOD and LOQ were in the range of 0.12-0.37 and 0.60-1.85 µg/zone, respectively. Linearity was found over the concentration range of 0.5-4.0 µg/zone for quercetin, kaempferol, sciadopitysin, ginkgetin, and bilobetin with correlation coefficients r = 0.9990, 0.9922, 0.9939, 0.9974, and 0.9706, respectively. Average recoveries were in the range of 97.7-100.4 %.

Baill with methanol - 25 % hydrochloric acid 4:1 and sonication for 1 h (these were the best conditions of five different solvent compositions and different sonication times investigated). TLC of the obtained extracts on silica gel with 1) petroleum ether (60-90 ºC) - acetone 5:2, detection by spraying with 10 % sulfuric acid in ethanol and heating at 105 ºC; 2) toluene - ethyl acetate - formic acid 5:2:1, detection by spraying with 1 % aluminium chloride in ethanol and evaluation under UV 366 nm; 3) hexane - ethyl acetate - formic acid 70:50:8, detection by spraying with 1 % aluminium chloride in ethanol and heating at 105 ºC, detection under daylight and UV 366 nm; 4) toluene - ethyl acetate - formic acid 5:4:1 saturated with hydrochloric acid, detection under daylight and UV 366 nm. System 4) provided the best separation and was most practical. Identification of quercetin by fingerprint comparison with the standard.

For quality control, TLC of the extracts of the medicine on silica gel 1) for Poria cocos, with petroleum ether (30-60 ºC) – acetone – ethyl acetate 85:15:1, detection at UV 366 nm; 2) for Licorice, with chloroform – methanol – water 40:10:1, detection by spraying with 10 % sulfuric acid in ethanol and heating at 105 ° until the zones are visible, evaluation in daylight.

      Chromatogr. 26 (1), 61–68 (2012) Picroside-I and picroside-II are known bioactive metabolites in Picrorhiza species. Presentation of a simple, precise method for the simultaneous determination of picrosides (picroside-I and picroside-II) in two different Picrorhiza species, P. kurroa and P. scrophulariiflora. TLC of the extracts of the medicinal herbal drugs on silica gel with chloroform – methanol 22:3. Quantification of picrosides (picroside-I and picroside-II) by absorbance measurement at UV 254 nm. Comparative study revealed that the content of picroside-I and picroside-II is higher in P. scrophulariiflora than P. kurroa the content of picroside-I was found to be 1.3 and 1.6 % inP. kurroa and P. scrophulariiflora, and of picroside-II 0.5 and 0.6 %, respectively. Study of the antioxidant potential of the two Picrorhiza species using DPPH* radical reagent. The scavenging activities of P. kurroa and P. scrophulariiflora were 37.7 % and 34.3 %, respectively, at a concentration of 0.1 mg/mL.

For quality control, TLC on silica gel 1) for Radix Angelicae Pubescentis, with cyclohexane – ethyl acetate – formic acid – glacial acetic acid 70:20:2:3, detection under UV 366 nm; 2) for Radix Notoginseng and Radix Dipsaci, with n-butanol – ethyl acetate – 10 % diluted ammonia 4:1:5, detection by spraying with 10 % sulfuric acid in ethanol and heating at 105 °C, viewing in daylight; 3) Herba Taxilli, with toluene saturated with water – ethyl formate – formic acid 5:4:1, detection by spraying with 5 % aluminium chloride in ethanol and heating at 105 °C, viewing under UV 366 nm. Quantification of osthole by HPLC.
