Bioautography by dipping into a suspension of Bacillus subtilis for 8 s, followed by incubation for 17 h at 37 °C. The plates were sprayed with 0.2 % aqueous solution of 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) and incubated for 30 min at 37 °C. Thymus citriodorus and Thymus marschallianus were selected as promising targets for further investigations.

Detection by exposure to iodine vapor. The hRF values for (1) to (4) were 52, 15, 7 and 10, respectively.

      Determination of the hemolytic activity of saponins by an HPTLC-blood gelatin test

HPTLC of steroid alkaloid saponins from potato sprouts on silica gel with chloroform – methanol – water 60:40:9 with chamber saturation without filter paper over a migration distance of 70 mm. Detection by dipping half of the plate in anisaldehyde-sulfuric acid reagent, drying for 10 min and heating at 70 °C for 5 min. The other half of the plate was used for the blood-gelatin test: dipping in 0.5 % polyisobutylmethacrylate in n-hexane – chloroform 20:1 for 10 s, coating with 40 mL blood-gelatin (2 % gelatin in PBS buffer pH 7.4 with 3 % human blood preservation), storage for 12-48 h at 4 °C. Evaluation under UV 366 nm and white light. Semiquantitative evaluation of the hemolytic activity using the substance digitonin from potato sprouts. The hemolytic activity of α-chaconine was comparable to the effect of 16-20 µg digitonin.

      Anticonvulsant activity and acute neurotoxic profile of Achyranthes aspera Linn

      J. Ethnopharmacol. 202, 97-102 (2017). HPTLC of betaine in the roots of Achyranthes aspera on silica gel with methanol – water 9:1. Detection by spraying with Dragendorff's reagent, followed by 10 % ethanolic sulfuric acid and drying at 110 °C for 5 min. Quantitative determination by densitometry at 520 nm. The hRF value for betaine was 36. Linearity was between 1 and 5 μg/band. The intermediate precision was below 2 % (n=6). LOD and LOQ were 0.13 μg/mL and 0.10 μg/mL, respectively. (Note: The reported LOD and LOQ do not make sense, as LOD is higher than LOQ.)
