      Simultaneous quantification of flavonoids in Ginkgo biloba using RP-HPTLC densitometry method

HPTLC of flavonoids (quercetin and kaempferol) and biflavonoids (sciadopitysin, ginkgetin, and bilobetin) in aqueous methanolic extracts of Ginkgo biloba on RP-18 by double development with 1) acetonitrile - water - methanol - formic acid 20:20:1:0.005 and 2) acetonitrile - water - methanol - formic acid 20:17:1:0.005. Quantitative determination by densitometry in absorption mode at 254 nm. LOD and LOQ were in the range of 0.12-0.37 and 0.60-1.85 µg/zone, respectively. Linearity was found over the concentration range of 0.5-4.0 µg/zone for quercetin, kaempferol, sciadopitysin, ginkgetin, and bilobetin with correlation coefficients r = 0.9990, 0.9922, 0.9939, 0.9974, and 0.9706, respectively. Average recoveries were in the range of 97.7-100.4 %.

      A marker-based stability indicating high-performance thin-layer chromatography method for Vitex trifolia

      J. Liq. Chromatogr. Relat. Technol. 34, 1925-1937 (2011). HPTLC of p-hydroxy benzoic acid (1), chrysoplenol-D (2), p-methoxy benzoic acid (3) and casticin (4) in the aerial parts of Vitex trifolia on silica gel with chloroform - methanol 24:1 + 1 drop formic acid. Quantitative determination by absorbance measurement at 254 nm. The hRf values of compounds (1) - (4) were 25, 31, 63 and 87, respectively. LOD and LOQ were found to be 18-58 ng/zone for (1), 39-132 ng/zone for (2),15-52 ng/zone for (3) and 33-111 ng/zone for (4). Repeatability and reproducibility (%RSD) for (1)-(4) were found in the range of 0.8-1.2 % and 1.2-1.3 %, respectively. Recoveries were obtained in the range of 94.0-101.1 %, 97.8-102.0 %, 95.1-100.1 %, and 97.6-100.3 % for compounds (1) - (4), respectively.

      HPTLC quantification and antimicrobial activity of ursolic acid from Diospyros melanoxylon

Detection by dipping in 5 % methanolic sulphuric acid reagent and heating at 105 °C for 3 min. Quantitative determination by absorbance measurement at 540 nm. Linearity was in the range of 50-450 ng/zone for ursolic acid. Limits of detection and quantification were found to be 20 and 40 ng/zone. Recovery (by standard addition) was 97.5 %.

Quantitative determination by absorbance measurement at 240 nm. The hRf of compounds (1) and (2) were 30 and 48, respectively. Linearity was in the range of 200-1000 ng/zone. Limits of detection and quantification were found to be 13 and 22 ng/zone for (1) and 18 and 31 ng/zone for (2), respectively. Recovery was found to be 99.2-99.5 % for (1) and 98.9-99.2 % for (2), respectively. The method showed comparable results to a validated HPLC method.

Baizhuhugan compound oral liquid is effective for enriching the blood, tonifying spleen, soothing the liver, and is clinically prescribed to cure chronic hepatitis and cirrhosis. For quality control, identification of the three contained herbal drugs by TLC of the extracts of the preparation 1) for Chinese Angelica, on silica gel with n-hexane – ethyl acetate 9:1, detection at UV 366 nm; 2) for Bighead Atractylodes rhizome, on silica gel with cyclohexane – ethyl acetate 7:3, detection by spraying with 10 % sulfuric acid in ethanol and heating at 105 °C, viewing in daylight; 3) for Chinese herbaceous Peony, on silica gel with chloroform - ethyl acetate – methanol – ammonia 8:1:4:1, detection by spraying with 5 % vanillin in ethanol – sulfuric acid 4:1 and heating mildly, viewing in daylight.

Recovery was found to be 98.2-101.4 % for (1), 98.0-101.1 % for (2), 98.2-100.5 % for (3) and 98.1-101.9 % for (4).

Miq, with toluene – propanone – methanol 16:6:1 after preconditioning with ammonia vapors, detection by spraying with 5 % potassium iodobismuthate and viewing in daylight.
