      Effects of high and low temperatures on the lipid content of the digestive gland gonad complex of Biomphalaria glabrata as determined by high-performance thin-layer chromatography

HPTLC of neutral and polar lipid content of digestive gland-gonad complex of Biomphalaria glabrata on silica gel with petroleum ether - diethyl ether - glacial acetic acid 80:20:1 for neutral lipids, and chloroform - methanol - deionized water 65:25:4 for phospholipids. Detection of neutral lipids by spraying with 5 % ethanolic phosphoric acid, followed by heating at 115 ºC for 10 min. Phospholipids were detected by spraying with 10 % cupric sulfate in 8 % phosphoric acid, followed by heating at 140 ºC for 30 min. Quantitative determination by absorbance measurement at 610 nm for neutral lipids and 370 nm for polar lipids. The hRF values of neutral lipids were 19 for cholesterol, 38 for oleic acid and 57 for triolein, whereas for polar lipids were 46 for phosphatidylcholine and 66 for phosphatidylethanolamine.

      Application of image analysis technique for the determination of organophosphorus pesticides by thin-layer chromatography

      J. Planar Chromatogr. 29, 221-226 (2016). HPTLC of malathion (1) and fenthion (2) in apple and orange juice on silica gel with n-hexane – acetone 3:2. Detection by spraying with 6 % sodium azide, 0.25 % starch, pH 6.5 for (1) and 0.5 % sodium azide, 2 % starch,_x000D_ pH 6.0 for (2), followed by exposure to iodine vapors for 15 s. The chromatograms were scanned at 300 dpi. The hRF values for (1) and (2) were 51 and 48, respectively. Linearity was in the range of 1.0-7.3 μg/zone for (1) and 2.5-24.8 μg/zone for (2). Intermediate precisions were below 3.1 %. The LOD and LOQ were 242 and 807 ng/zone for (1) and 588 and 1960 ng/zone for (2), respectively. Recovery was in the range of 94.8-105.4 % for (1) and 84.2-104.0 % for (2).

      Unprecedented sensitivity of the planar yeast estrogen screen by using a spray-on technology

Development of a new spray-on method for applying yeast cells to HPTLC plates, leading to a much higher sensitivity of the planar yeast estrogen screen (p-YES), which can serve as a highly valuable and sensitive screening tool for the detection of estrogenic compounds in various sample matrices such as water and wastewater, personal care products and foodstuff. HPTLC of sample constituents and direct detection of estrogenic compounds by spraying with yeast cells. This resulted in much sharper signals compared to those in previous publications. Satisfying results were achieved using cultures with cell densities of 1000 FAU with reduced signal broadening, thus lower LOQ for estrogenic compounds, e.g. estrone 2 pg/zone, 17β-estradiol 0.5 pg/zone, 17α-ethinylestradiol 0.5 pg/zone and estriol 20 pg/zone. Demonstration of the possibility of the method to characterize profiles of estrogenic activity of wastewater samples with high quality and reproducibility by using native samples from wastewater or even surface water directly applied on HPTLC plates without the need for prior sample treatment.
