      Quantitative determination of the main hydroxyanthaquinones in soils by TLC with two-stage development

      J. Planar Chromatogr. 9, 48-51 (1996). TLC of 7,7'-biphysicon, physicon, chrysophanol on silica with chloroform - ethyl acetate - formic acid 89:10:1. After cutting the plate into two pieces 7.0 cm from the bottom edge, the upper portion was developed with the same solvent for 1.0 cm, and after drying with hexane - ethyl acetate - formic acid - water 178:20:2:7. Quantification by densitometry at 430 nm and a reference wavelength of 540 nm. Detection limit in the lower nanogram range (5 - 10 ng).

      Analysis of polycyclic aromatic hydrocarbons

      CBS 83, 6-7 (1999). HPTLC-AMD of PAHs in liver and lung of animals exposed to contaminated soil on RP-18 with a 14-step gradient from acetonitrile to methanol - water 9:1. Quantification by fluorescence measurement at 313/>400 nm and 254/>400 nm, respectively. Strict linearity (r2 > 0.99 on 5 measuring points) was proven. Coefficient of variation was found to be < 5 %, and recoveries between 55 and 70 % were given. Limits of detection was determined to be 0.2 to 2 ng/g sample.

HPTLC of pesticides (prochloraz, chloroxuron, chlorotoluron, metoxuron, chlorbromuron, cyanazine, flamprop-M-isopropyl, prometryn, permethrin, bitertanol, hexazinone, chlorsulfuron, methabenzthiazuron, phenmedipham, metiocarb, dichlofluanid, propachlor, procymidone, terbuthylazine, propyzamide, tri-allate, bifenthrin) on silica gel, RP-18 W, RP-18, and cyano phases in a horizontal chamber. In adsorbent-gradient TLC silica gel plates were used in the first direction with tetrahydrofuran - n-heptane 1:4. After drying, the plates were cut in narrow strips and connected with RP-18 W or cyano plates, the pesticides were transferred to the before mentioned plates with methanol and developed in the second dimension with water - methanol or water - dioxane. Detection under UV light at 254 or 366 nm. Quantitative determination by densitometry at 254 nm.

      Two new methods for early detection of the effects of herbicides in plants using biomarkers

Presentation of two simple and rapid HPTLC methods for early detection of the effects of herbicides using two different groups of plant biomarkers, which were developed as field tests (Herbicide Weed Response test - HWR-Test). Phytochemical changes can be detected before any morphological changes are visible on the plants. These changes are defined as biomarkers and can be detected by HPTLC-screening. After overall identification of the phytochemical biomarker pattern, two different biomarker groups, carbohydrates and amino acids, were detected using modified reagents for color reactions. Evaluation under daylight and videodensitometric analysis of digital images by VideoScan software. The screening method was previously described [H. W. Ravn, M. Hjorth, L. Lauridsen, P. Kudsk, S. K. Mathiassen, L. Mondolot, Bull. Environ. Contam. Toxicol. 75, 236-245 (2005)].

      TLC-bioautography analysis of vitamin B12 compound from the short-necked clam (Ruditapes philippinarum) extract used as a flavoring

TLC of vitamin B12 on silica gel with 2-propanol - 28 % ammonia - water 7:1:2 and 1-butanol - 2-propanol - water 10:7:10 in the dark at room temperature. After drying, agar containing basal medium and pre-cultured E. coli 215 was overlaid and then incubated at 30 °C for 20 h. Detection by spraying with a methanolic solution of 2,3,5-triphenyltetrazolium salt, B12 compounds appeared as red zones indicating E. coli growth.

      Chinese J. of Inform. on TCM 20 (7), 52-54 (2013). Yangxue Yishen capsules are a herbal TCM preparation for treating rough skin, blood scale disease, alopecia areata and neurodermatitis. For quality control, TLC on silica gel 1) for Gastrodia elata Blume and the standard gastrodine, with chloroform – ethyl acetate – methanol – formic acid 80:10:30:1, detection by spraying with 10 % phosphomolybdic acid in ethanol and heating at 110 °C until the zones are visible in daylight; 2) for Epimedium davidii Franch and the standard icariin, with benzene – methanol – glacial acetic acid 45:8:4, detection by spraying with 10 % aluminium chloride in ethanol and letting stand for 40 min at room temperature, evaluation under UV 366 nm; 3) for Salvia miltiorrhiza and the standard protocatechuic aldehyde, with chloroform – acetone – formic acid 25:10:4, detection by exposure to ammonia vapors for 15 min and viewing under UV 254 nm. Quantification of astragaloside A by HPLC-ELSD.

      Application of image analysis technique for the determination of organophosphorus pesticides by thin-layer chromatography

Detection by spraying with 6 % sodium azide, 0.25 % starch, pH 6.5 for (1) and 0.5 % sodium azide, 2 % starch,_x000D_ pH 6.0 for (2), followed by exposure to iodine vapors for 15 s. The chromatograms were scanned at 300 dpi. The hRF values for (1) and (2) were 51 and 48, respectively. Linearity was in the range of 1.0-7.3 μg/zone for (1) and 2.5-24.8 μg/zone for (2). Intermediate precisions were below 3.1 %. The LOD and LOQ were 242 and 807 ng/zone for (1) and 588 and 1960 ng/zone for (2), respectively. Recovery was in the range of 94.8-105.4 % for (1) and 84.2-104.0 % for (2).

Improvement of a known TLC procedure regarding sensitivity.
