      Semiquantitative estimation of enrofloxacin and ciprofloxacin by thin-layer chromatography - direct bioautography

      J. Liq. Chrom. Rel. Technol. 27, 2071-2085 (2004). HPTLC of enrofloxacin and ciprofloxacin on silica gel using dichloromethane - methanol - 2-propanol - 25 % ammonia 3:3:5:2. Bioautography with Chrom Biodip® Antibiotics Test Kit. The developed and dried plate was immersed briefly in the microorganism solution and incubated overnight at 28 °C. After incubation the plate was sprayed with 3-[4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT)-solution and incubated for about 30 min. The dried plates were scanned for documentation. The areas of the inhibition zones were then measured with a planimeter.

      Analysis of glycosides and aglycones of flavonoid compounds by double-development thin-layer chromatography

HPTLC of aglycones and glycosides (flavone, hesperetin, naringenin, apigenin, kaempferol, quercetin, myricetin, tiliroside, apigenin 7-glucoside, myricitrin, kaempferol 3,7-dirhamnoside, hyperoside, hesperidin, rhoifolin, rutin, naringin) on silica gel by using mixtures of dichloromethane and ethyl acetate for aglycones and mixtures of ethyl acetate and formic acid for glycosides. The plates were conditioned for 15 min and developed face down in a horizontal chamber. To separate mixtures of flavonoids two-step gradient devlopment was used. In the first step (for gycosides) the plates were developed to a distance of 65 mm with ethyl acetate - formic acid - water 170:30:1. In the second step (for aglycones ) the plates were developed to a distance of 95 mm with dichloromethane - ethyl acetate - formic acid 170:30:1. Quantitation by densitometry at 254 nm.

The method was validated for accuracy, precision, linearity, specificity, LOD, and LOQ.

      Evaluation of N-substituted 2,4-dihydroxyphenylthioamide fungicide lipophilicity using the chromatographic techniques HPLC and HPTLC

      Chromatographia 62 (3-4), 121-126 (2005). 2,4-Dihydroxyphenylthioamide derivatives modified on the N-aryl ring have substantial fungicidal activity. To determine their quantitative structure-activity relationships their lipophilicity was determined by use of column liquid chromatography and thin-layer chromatography. Methanol - water systems were used as mobile phases and the linear dependences of retention (RM and log k) on volume fraction of organic modifier, phi, were determined. This enabled precise determination of lipophilicity (RMw and log kw) by extrapolation. Correlations were found between quantities characterizing the lipophilicity of the compounds. Deviations enabled discovery of compound structural features which increase or reduce lipophilicity. When these data were correlated with biological activity against the phytopathogenic fungi Alternaria alternata and Botrytis cinerea parabolic dependences were obtained.
