      High-Performance Thin-Layer Chromatographic– Densitometric determination of the major triterpene saponins and their aglycones from Centella asiatica

      J. Planar Chromatogr. 32, 25-30 (2019). HPTLC of asiaticoside (1) and madecassoside (2) in Centella asiatica on silica gel with chloroform - methanol - water 5:4:1. Quantitative determination by absorbance measurement at 202 nm. The hRF values for (1) and (2) were 72 and 47, respectively. Linearity was between 100 and 1500 ng/zone for both (1) and (2). The intermediate precision was below 4.3 % (n=3). The LOD and LOQ for (1) and (2) were 14 and 60 as well as 37 and 82 ng, respectively. Recovery rate was 103.3 % for (1) and 95.8 % for (2).

      Validated High-Performance Thin-Layer Chromatographic–Densitometric method for the isolation and standardization of ayapanin in Ayapana triplinervis

      J. Planar Chromatogr. 32, 41-46 (2019). HPTLC of ayapanin in the leaves of Ayapana triplinervis on silica gel with ether - ethyl acetate 3:2. Quantitative determination by absorbance measurement at 254 nm. The hRF value of ayapanin was 56. Linearity was between 200 and 1000 ng/zone. The intermediate precision was below 0.8 % (n=6). The LOD and LOQ for ayapanin were 54 and 169 ng/zone, respectively. Recovery rate was 99.7 % for ayapanin.

      J. Liq. Chromatogr. Relat. Technol. 37, 2784-2799 (2014). HPTLC of atorvastatin (1), ezetimibe (2) and fenofibrate (3) on silica gel with toluene – methanol – triethylamine 16:3:1. Quantitative determination by absorbance measurement at 245 nm. The hRF values for (1) to (3) were 10, 20 and 80, respectively. Linearity was in the range of 100-350 ng/zone for (1) and (2) and 1600-5600 ng/zone for (3). The intermediate/interday/intra-day precisions were below 1 % (n=3). The LOD and LOQ were 60 and 90 ng/zone for (1), 80 and 100 ng/zone for (2) and 90 and 150 ng/zone for (3), respectively. Recoveries were in the range of 97.6-102.6 %.

For quality control, TLC on silica gel 1) for Vaccaria segetalis, with chloroform – methanol – water 15:7:2, detection by spraying with fresh solution of (bismuth subnitrate - glacial acetic acid – water 17:200:800) – 40 % potassium iodate – glacial acetic acid – water 1:1:4:15, evaluation in daylight; 2) for Scrophularia ningpoensis Hemsl., with ethyl acetate - methanol – formic acid – glacial acetic acid - water 30:1:2:2:4, detection by spraying with 1 % vanillin in sulfuric acid – ethanol 1:4 and heating at 105 °C until the zones were visible in daylight; 3) for Angelica sinensis, with n-hexane – ethyl acetate 4:1, detection under UV 366 nm.

The intermediate intra-day and inter-day precisions were below 2.3 % (n=3). The LOD and LOQ were 530 and 1760 ng/zone for (1), 240 and 790 ng/zone for (2) and 110 and 370 ng/zone for (3), respectively. Recoveries were in the range of 98-105 % for (1), 101-131 % for (2) and 95-116 % for (3).

The intermediate intra-day and inter-day precisions were below 2.9 % (n=3). Recoveries were found in the range of 100.2-102.4 %.

For quality control, TLC on silica gel (1) for Cyperus rotundus, with petroleum ether (60-90 °C) – ethyl acetate 15:1, detection at UV 254 nm; (2) for Atractylodes macrocephala Koidz. and atractylone with petroleum ether (60-90 °C) – ethyl acetate 50:1, detection by spraying with 5 % vanillin in sulfuric acid – ethanol 1:4, followed by heating until zones are visible; (3) for Angelica dahurica (Fisch. ex Hoffm.) Benth. et Hook. f. ex Franch. et Sav. and imperatorin with petroleum ether (60-90 °C) – diethyl ether 1:1, detection at UV 366 nm; (4) for Epimedium brevicornum Maxim and icariin with ethyl acetate – butanone – formic acid – water 10:1:1:1, detection at UV 366 nm before and after spraying with 3 % aluminium trichloride in ethanol.
