Gongxueting Keli granules are a herbal TCM preparation for treating women’s asthenia of both the spleen and kidney, and blood stasis due to menstruation and metrorrhagia. For quality control, TLC on silica gel 1) for Eclipta and the standard ecliptasaponin A, with dichloromethane – ethyl acetate – methanol – water 30:40:15:3, detection by spraying with 10 % sulfuric acid in ethanol and heating at 105 °C until the zones are visible in daylight; 2) for Leonurus Artemisia (Laur.) S. Y. Hu F and the standard stachydrine hydrochloride, with acetone – ethanol – hydrochloric acid 10:6:1, detection by heating at 105 °C for 15 min followed by spraying with 10 % sulfuric acid in ethanol and then heating, followed by spraying with 5 % potassium indobismuthate – 1 % ferric chloride in ethanol 10:1, viewing in daylight.

      J. of China Pharm. 26 (6), 620-622 (2012). Rheum palmatum L., a herbal TCM drug for clearing away body heat, eliminating toxin and nourishing skin, is commonly used as a component of herbal TCM preparations. However, a homologous herb, Rumex madaio Makino (syn. Rumex daiwoo Makino) is sometimes mistaken for Rheum palmatum in drug production. For quality control of rhaponiticin in antibacterial Kangjun Xiaoyan Jiaonang capsules, TLC of the extracts and the standard rhaponiticin on silica gel with ethyl acetate – acetone – water 10:7:1, detection under UV 366 nm. Quantification of rhaponiticin by HPLC. The method was successfully applied to nine batches of real life samples from five pharmaceutical factories.

      Thin-layer chromatography–image analysis method for the simultaneous quantification of andrographolide and related diterpenoids in Andrographis paniculata

      J. Planar Chromatogr. 27, 140-144 (2014). HPTLC of andrographolide (1), neoandrographolide (2) and deoxyandrographiside (3) in the aerial parts of Andrographis paniculata on silica gel with dichloromethane - toluene - ethanol 13:5:3. Detection by dipping into p-anisaldehyde–sulfuric acid reagent, followed by heating at 110 ºC for 6 min. For quantification, image analysis with an office scanner at 200 dpi and densitometric measurement in absorbance mode at 540 nm. The hRF values for (1) to (3) were 61, 35 and 16, respectively. Linearity was in the range of 3-24 μg/zone for (1) and 1-8 μg/zone for (2) and (3). The intermediate/interday/intra-day precisions were below 3 % (n=3). The LOD and LOQ were 510 and 1700 ng/zone for (1), 140 and 480 ng/zone for (2) and 6 and 190 ng/zone for (3), respectively. Recovery was between 96.1 and 104.3 % for (1) to (3). Comparable results were obtained with the TLC-image analysis method, TLC-densitomety and HPLC.

The hRF value for clotrimazole was 27. Linearity was in the range of 1000-2400 ng/zone. The intermediate/interday/intra-day precisions were below 1.3 % (n=6). Recovery was between 99.1 and 100.1 %.

For quality control, TLC on silica gel 1) for Gardenia jasminoides Ellis, with ethyl acetate – acetone – formic acid – water 5:5:1:1, detection by spraying with 10 % sulfuric acid in ethanol and heating at 110 °C until the zones are clearly visible in daylight; 2) for flowers and buds of Sophora japonica L., with ethyl acetate –formic acid – water 8:1:1, detection after spraying with 1 % aluminium chloride in ethanol under UV 366 nm; 3) for Paeonia suffruticosa Andr., with cyclohexane - ethyl acetate 3:1, detection by spraying with 2 % ferric chloride in ethanol and heating mildly until the zones are clearly visible in daylight. Quantification of rutinum by HPLC.

Bunge and the standard astragaloside IV, with chloroform – methanol – water 13:7:2, detection by spraying with 10 % sulfuric acid in ethanol and heating at 105 °C until the zones are visible in daylight; 3) for Lignum sappan, with chloroform – acetone – formic acid 14:4:1, detection a) in daylight after letting the plates stand for 6 hours, b) under UV 366 nm; 4) for Radix bupleuri and the standard saikosaponin A, with ethyl acetate – ethanol – water 8:2:1, detection by spraying with 2 % p-diethylaminobenzaldehyde in 40 % sulfuric acid and heating at 60 °C until the zones are visible in daylight.

Choisy, with toluene – glacial acetic acid 100:1, detection by spraying with 5 % vanillin in sulfuric acid – ethyl acetate 22:1 and heating mildly until the zones are visible in daylight; 3) for Amomum microcarpum C. F. Liang et D. Fang and Amomum kravanh, with cyclohexane – glacial acetic acid 100:1, detection by spraying with 5 % vanillin in sulfuric acid – ethanol 1:4 and heating mildly until the zones are visible in daylight. Quantification of aloe emodin by HPLC.

Quantitative determination by absorbance measurement at 246 nm. The hRF value of swertiamarin was 58. Linearity was between 200 and 1200 ng/zone. The intermediate intra-day and inter-day precisions were below 2 % (n=6). The LOD and LOQ were 38 and 115 ng/zone, respectively. Recoveries were in the range of 98-100 %.
