      A stability indicating HPTLC method for the analysis of irinotecan in bulk drug and marketed injectables

      J. Liq. Chromatogr. Relat. Technol. 34, 1502-1517 (2011). HPTLC of irinotecan in bulk drug and injectable formulations on silica gel with acetone - ethyl acetate 17:3 + 1 drop acetic acid. Quantitative determination by absorbance measurement at 366 nm. The hRf of irinotecan was 31. Linearity was 50-500 ng/zone. The LOD and LOQ was found to be 10 and 33 ng/zone. The intra-day precision was 4.3 % (n = 6) and inter-day precision over three different days was 3.1 %. Intra-day and inter-day accuracy were 96.9-100.3 % and 96.5-98.7 %, respectively. Recovery (by standard addition) ranged from 94.6-101.4 %.

      Simultaneous quantification of flavonoids in Ginkgo biloba using RP-HPTLC densitometry method

HPTLC of flavonoids (quercetin and kaempferol) and biflavonoids (sciadopitysin, ginkgetin, and bilobetin) in aqueous methanolic extracts of Ginkgo biloba on RP-18 by double development with 1) acetonitrile - water - methanol - formic acid 20:20:1:0.005 and 2) acetonitrile - water - methanol - formic acid 20:17:1:0.005. Quantitative determination by densitometry in absorption mode at 254 nm. LOD and LOQ were in the range of 0.12-0.37 and 0.60-1.85 µg/zone, respectively. Linearity was found over the concentration range of 0.5-4.0 µg/zone for quercetin, kaempferol, sciadopitysin, ginkgetin, and bilobetin with correlation coefficients r = 0.9990, 0.9922, 0.9939, 0.9974, and 0.9706, respectively. Average recoveries were in the range of 97.7-100.4 %.

      Method validation of marmelosin from fruit pulp of Aegle marmelos (L) correa using HPTLC technique

Densitometric quantification of marmelosin at 310 nm. The method was linear in the range of 50-350 ng/band. The method was used to determine the marmelosin content of pulp, unripe pulp, seeds, leaves, rind, outer stain and inner stain.

      Liquid chromatographic resolution of the enantiomers of metoprolol and carvedilol in pharmaceutical formulations by use of Marfey’s reagent and its variants

TLC of (R,S)-metoprolol and (R,S)-carvedilol, derivatized with 1-fluoro-2,4-dinitrophenyl-5-L-alanine amide (Marfey’s reagent, FDNP-L-Ala-NH2) and its six structural variants (FDNP-L-Phe-NH2, FDNP-L-Val-NH2, FDNP-L-Pro-NH2, FDNP-L-Leu-NH2, FDNP-L-Met-NH2, and FDNP-D-Phg-NH2), on RP-18 in a saturated chamber at 25 +/- 2 °C. The best separation was obtained with TEAP buffer (pH 5.5, 50 mM) - acetonitrile 1:1. The LOD for metoprolol and carvediol were in the range of 240 to 350 ng/zone for each enantiomer. The intra-day and inter-day precision (%RSD) was between 0.1-0.2 for (S)-metoprolol and 0.2-0.8 for (R)-metoprolol.

      TLC-densitometric determination of tolperisone and its impurities 4-methylpropiophenone and piperidine in pharmaceutical preparations

HPTLC of tolperisone (1) and its impurities 4-methylpropiophenone (2) and piperidine (3) on silica gel with cyclohexane-1,4 - dioxane - isopropanol - ethanol 32:1:2:8 + 1 drop glacial acetic acid. Detection by dipping in a 0.3 % methanolic ninhydrin solution for 10 min, followed by heating at 105 °C for 5 min. Quantitative determination by absorbance measurement at 570 nm. The hRf values of compounds (1) to (3) were 10, 76 and 60, respectively. Linearity was in the range of 60-1500 ng/band for (1), 90-400 ng/band for (2) and 40-250 ng/band for (3). Limits of detection and quantification were 20 and 60 ng/band for (1), 30 and 90 ng/band for (2) and 20 and 40 ng/band for (3), respectively.The intermediate precisions (level 2) for (1) to (3) were 1.6 %, 2.6 % and 2.4 % (n=5), respectively. Recovery for compounds (1) to (3) was between 84.6 and 99.7 %.

For quality control, TLC on silica gel 1) for Cortex Phellodendri Chinensis with n-butanol – glacial acetic acid – water 9:2:1, detection by viewing at UV 366 nm, identification by fingerprint comparison in parallel with the standard drug and berberine chloride standard submitted to the same procedure; 2) for Flos Lonicerae and Fructus Forsythiae with the upper phase of ethyl acetate – formic acid – water 14:5:5, detection by viewing at UV 366 nm, identification by fingerprint comparison in parallel with the standard drug and chlorogenic acid standard submitted to the same procedure.

TLC on silica gel with petroleum ether (60-90 ºC) – ethyl acetate 50:1, detection by spraying with 5 % vanillin in a solution of ethanol – sulfuric acid 100:1 and heating at 105 °C until the zones were visualized.

TLC of the extracts and standards resveratrol and bergeninum on silica gel with chloroform – ethyl acetate – formic acid 30:20:1, detection by spraying with a solution of vanillin - ethanol - sulfuric acid 10:200:1.
