Staining of textiles often uses several dyes matching in color, and a change in the dye concentration of the staining solution influences the dye uptake rate in the dyeing process, and thus affects the dyeing result. The real-time monitoring of the dye concentration in the dyeing process can reflect the state of dyeing and allows for more reasonable working processes and staining formulas. A TLC method is presented for multi-wavelength scanning of the dyes cationic yellow X-5GL and X-8GL on silica gel without the need for chromatogram development. An oblique projection algorithm was based on the oblique projection theory. This theory describes a multi variable system with data space and establishes a pure signal separation model which allows to differentiate the measured vector spectrum signal of the interested component from the mixed spectrum signal and determine its content by chemometrics. The obtained results were compared with those by using conventional TLC on silica gel developed with ethanol, followed by densitometric evaluation. It was found that the measured precision for the presented method was lower than that of traditional TLC.

For quality control, TLC on silica gel 1) for Angelica sinensis and Ligusticum chuanxiong Hort, with n-hexane – ethyl acetate 19:1, detection at UV 366 nm; 2) for Atractylodes lancea (Thunb.) DC., with petroleum ether (60-90 °C) – ethyl acetate 19:1, detection by spraying with 5 % p-dimethylaminobenzaldehyde in sulfuric acid – ethanol 1:4 and evaluation under white light; 3) for Corydalis yanhusuo W. T. Wang ex Z. Y. Su et C. Y. Wu and the standard tetrahydropalmatine, with n-hexane – chloroform – methanol – triethylamine 10:6:1:1, detection by exposure to iodine vapors until the zones are clearly visualized, evaluation under white and at UV 366 nm; (4) for Pericarpium Citri reticulatae and the standard hesperidin, with chloroform – acetone – methanol 5:1:1, detection by spraying with 3 % aluminium trichloride in ethanol and heating mildly until the zones are visible under white and at UV 366 nm. Jinwu Gutong Jiaonang capsules are a TCM preparation for nourishing liver and kidney, activating blood, prescribed to treat rheumatic arthralgia, osteoporosis, lumbago and limb numbness, impotence etc. For quality control, HPTLC of icariin on silica gel with chloroform – methanol – water 27:12:4, detection by spraying with 10 % sulfuric acid in ethanol and heating at 105 °C until the zones are visible.

The hRF value of erythrocentaurin was 54. Linearity was in the range of 200-1500 ng/zone. LOD and LOQ were 60 and 180 ng/zone, respectively. Intra-day and inter-day precision was below 1 % (n=3). Recoveries were between 97 and 99 %.

Quantitation by absorbance measurement at UV 366 nm. Linearity was between 50 and 300 ng/zone. The LOD and LOQ for ochratoxin A was 10 and 30 ng/zone, respectively. Recovery was in the range of 86-113 %.

Detection of (3) and (4) by spraying with stannous chloride 100 g/L in diluted hydrochloric acid, followed by heating at 100 °C for 1 min. Identification under UV light at 365 nm for (3) and (4). The hRF values for (1) to (4) were 40, 50, 46 and 54, respectively.

Quantitative determination by absorbance measurement at 265 nm. The hRF value for trigonelline was 19. Linearity was in the range of 100-2000 ng/zone. LOD and LOQ were 2 and 7 ng/zone. The intermediate precision was below 0.8 % (n=3). Average recovery was 99 %. Comparable results were obtained with a validated HPLC method.

The hRF value for pirfenidone was 49. Linearity was in the range of 800-1600 ng/zone. LOD and LOQ were 0.550 and 1.667 ng/zone. The intermediate precision was below 1.8 % (n=3). Recovery ranged between 100 and 102 %.
