      Determination of amikacin in parenteral dosage forms by high-performance thin-layer chromatography

HPTLC of amikacin (6-O-(3-amino-3-deoxy-a-D-glucopyranosyl) 4-O-(6-amino-6-deoxy-a-D-glucopyranosyl)-1-N-[(2S)-4-amino-2-hydroxybutyryl]-2-deoxy-D-streptamine) on silica with methanol - 25% NH3 - chloroform 10:10:1. After drying derivatization by immersion into 0.5% ninhydrin in n-butanol for exactly 1 s. Quantification by densitometry at 498 nm. Limit of detection and limit of quantification were 200 ng and 800 ng, respectively. RSD 1.22-1.49%.

Validation of the method by investigation of the procedures, including sample preparation, mobile phase, visualization, scanning wavelength, linearity range (0.28-1.65 µg, r=0.9991), precision (RSD <1.4 % within plate and <2.4 % plate-to-plate), resolution, repeatability, reproducibility, and recovery (87.2 %-99.7 % for different medicine). Comparison of the results with those obtained by HPLC.
