      Detection and determination of caffeine, taurine, and arginine in shampoos

      CBS 111, 5-6 (2013). HPTLC of caffeine, taurine, and arginine in shampoo samples extracted with isopropanol, on silica gel over 50 mm with isopropanol - n-heptane - water 7:3:1 for caffeine and isopropanol - water 4:1 for arginine and taurine. Detection under UV 254 nm (caffeine) and after spraying with ninhydrin reagent under white light (arginine and taurine). Quantitative absorbance measurement at UV 254 nm for caffeine and UV 600 nm for arginine and taurine. The hRF of caffeine was 54. Precision (%RSD) for the polynomial calibration of caffeine was 3.9 % (n=3). The hRf of taurine was 24. Arginine remained at the start position under these conditions. The content of taurine and caffeine found in shampoos corresponded to the usual amount of 0.1 % active ingredient in a formulation.

Mey. and the standards ginsenoside Rg1, Re, and Rb1 on silica gel with chloroform – methanol 4:1, detection by spraying with 10 % sulfuric acid in ethanol and heating at 105 °C until the zones are visible in daylight and under UV 366 nm; 3) for Ligusticum chuanxiong Hort. and the standards ligustrazine and ferulic acid on silica gel with n-hexane – ethyl acetate 9:1, detection under UV 366 nm; 4) for Paeonia veitchii and the standard paeoniflorin, on silica gel with chloroform – ethyl acetate – methanol – formic acid 200:25:50:1, detection by spraying with 5 % vanillin in sulfuric acid – ethanol 1:4 and heating at 105 °C until the zones are visible in daylight; 5) for Ephedra sinica Stapf and the standard (1R,2S)-(-)-ephedrine hydrochloride, on silica gel with chloroform – methanol – concentrated ammonia 40:10:1, detection by spraying with 0.1 % ninhydrin in acetone, heating at 105 °C until the zones are visible in daylight; 6) for Scutellaria baicalensis Georgi and standard baicalin, on plates made of silica gel - sodium acetate – 0.3 % CMC 50:8:150, with ethyl acetate – butanone – formic acid – water 5:3:1:1, detection by spraying with 2 % ferric chloride in ethanol, evaluation in daylight.

Zhiqiao Jianwei Keli granule is a herbal TCM for treatment of stagnated heat of liver and stomach, stomach pain and stomach cramps etc. For quality control, TLC on silica gel 1) for Fructus aurantii, with the lower phase of chloroform – methanol – water 13:6:2, detection by spraying with 3 % aluminum trichloride in ethanol and heating at 105 °C for 5 min and evaluation under UV 366 nm; 2) for Corydalis yanhusuo W. T. Wang ex Z. Y. Su et C. Y. Wu, with toluene – acetone 9:2, detection by exposure to iodine vapors for 3 min and evaluation under UV 366 nm; 3) for Forsythia suspensa (Thunb.) Vahl, with chloroform – methanol – glacial acetic acid 17:2:1, detection by spraying with 5 % vanillin in sulfuric acid – ethanol 1:4 and heating at 105 °C until the zones are visible in daylight. For determination of tetrahydropalmatine, TLC on silica gel with toluene – acetone 9:2 with chamber saturation for 30 min, detection by exposure to iodine vapors for 3 min. Evaluation under UV 366 nm and quantification by densitometric measurement at 343 nm using external standard calibration. Validation by investigating the linearity range of the calibration curve (55-330 ng/zone, r=0.996, n=6), the stability within 6 hours (%RSD 1.9 %, n=7), the precision (%RSD 1.3 %, n=6), and the repeatability (%RSD 0.8 %, n=6). Recovery was 98.0 % (%RSD 1.9 %, n=5).

      Pharmazie 68, 534-540 (2013). Detection and quantification by instrumental HPTLC and 1H-qNMR analyses. HPTLC of stachydrine on silica gel with methanol - dichloromethane - 25 % ammonia 8:2:3 with chamber saturation for 10 min in the horizontal developing chamber. The developing distance was 8.5 cm. Derivatization by dipping in Vagujfalvi solution (Wagner et al. 1983). Stachydrine was detected as red zone on yellow background. Quantitative determination by absorbance measurement at 517 nm. The method was selective with no other Dragendorff-positive constituents detected between hRF 45 - 60. The LOD was below 250 ng/zone. The results for the stachydrine content in the samples evaluated via peak height and peak area were in high accordance with independently performed 1H-qNMR measurements.

DC., with petroleum ether (60-90 °C) – ethyl acetate 19:1, detection by spraying with 5 % p-dimethylaminobenzaldehyde in sulfuric acid – ethanol 1:4 and evaluation under white light; 3) for Corydalis yanhusuo W. T. Wang ex Z. Y. Su et C. Y. Wu and the standard tetrahydropalmatine, with n-hexane – chloroform – methanol – triethylamine 10:6:1:1, detection by exposure to iodine vapors until the zones are clearly visualized, evaluation under white and at UV 366 nm; (4) for Pericarpium Citri reticulatae and the standard hesperidin, with chloroform – acetone – methanol 5:1:1, detection by spraying with 3 % aluminium trichloride in ethanol and heating mildly until the zones are visible under white and at UV 366 nm. Quantification of hesperidin by HPLC.

TLC of (1) chlorogenic acid and (2) caffeine in coffee samples on silica gel with ethyl acetate - methanol - water 77:13:10 to a distance of 8 cm using a horizontal sandwich chamber. The plates were dried at room temperature for 1 h. Detection of polyphenols with NP-PEG reagent, of terpenoids with anisaldehyde-sulfuric acid reagent followed by heating at 110 °C for 5 min, of purine derivatives with iodine-hydrochloric acid reagent. Quantitative determination of (1) and (2) at UV 254 nm using VideoScan software. The hRF value of (1) was 14, and of (2) 51. The antibacterial activity of green and roasted coffee seeds and pomace was evaluated against Bacillus subtilis using TLC-direct bioautography. TLC-2,2-diphenyl-1-picrylhydrazyl (DPPH) test was used to determine the antioxidant properties. For bioautography with Bacillus subtilis, the plates were immersed for 8 s in the bacterial suspension, placed in a moistened plastic box, and incubated at 37 °C for 17 h. Detection after spraying with 0.2 % MTT aqueous solution. For the DPPH test, plates were sprayed with 0.2 % methanolic DPPH solution. Detection of antioxidant activities as yellow zones against a purple background.
