      Ethyl acetate – propionic acid as green mobile phase system for selective densitometric identification of maltose with preliminary separation on silica high-performance thin-layer chromatographic plates

      J. Planar Chromatogr. 27, 19-22 (2014). HPTLC of maltose and fructose, dextrose, galactose and mannose on silica gel with ethyl acetate - propionic acid 1:1. Detection by dipping into orcinol solution (250 mg orcinol, 7.5 mL conc. sulfuric acid, 200 mL ethanol, and 50 mL water), followed by heating at 110 ºC for 5-10 min. Quantitative determination by absorbance measurement. The LOD for fructose, dextrose and mannose was 8 μg/zone and for maltose and galactose 2 μg/zone. Selective separation of maltose from other sugars was achieved using a mobile phase more suitable for the environment.

Xiaochaihutang Peifangkeli granule is a herbal TCM developed based on the classical Xiaochaihutang decoction prescription and is used for treatment of children's anorexia and vomiting, typhoid fever, malaria, jaundice, women’s hyperemesis gravidarum, and infections or inflammatory diseases. For quality control, TLC on silica gel 1) for Radix codonopsis, with toluene – ethyl acetate – formic acid – water 140:40:5:1, detection under UV 254 nm and by spraying with 10 % sulfuric acid in ethanol and heating at 105 °C until the zones are visible in daylight; 2) for Glycyrrhiza uralensis Fisch., with chloroform – methanol – water 40:10:1, detection by spraying with 5 % vanillin in sulfuric acid – ethanol 1:4 and heating at 105 °C until the zones are visible in daylight and under UV 366 nm. Quantification of baicalin by HPLC.

Detection by spraying with vanillin reagent (1 % in sulfuric acid), followed by heating at 105 ºC for 3 min. Quantitative determination by absorbance measurement at 365 nm. The hRf value for myotonin was 19. The intermediate/interday/intra-day precisions were below 3.4 % (n=5).

Bunge and the standard astragaloside A, with the lower phase of chloroform – methanol – water 13:7:2, detection under UV 366 nm and in daylight after spraying with 10 % sulfuric acid in ethanol and heating at 105 °C until the zones are visible. Quantification of naringin by HPLC.

Miq., Morus alba L. and Cynanchum glaucescens (Decne.) Hand.-Mazz on polyamide thin-films with (SDS - n-butanol – n-heptane 2.7:6.3:1.0) – formic acid 9:1, detection at UV 366 nm. Demonstration by applying to real life samples indicated that the procedure was suited for quality control.

Bunge. and astragaloside A with ethyl acetate – butanone – formic acid – water 5:3:1:1, detection by spraying with 10 % sulfuric acid in ethanol and heating at 105 °C for 5 min, evaluation under white light and at UV 366 nm; (2) for Atractylodes macrocephala Koidz. with cyclohexane - ethyl acetate 7:3, detection by spraying with 5 % p-dimethylaminobenzaldehyde in sulfuric acid – ethanol 1:4 and heating at 105 °C for 5 min, evaluation at UV 366 nm; (3) for Glycyrrhiza uralensis Fisch. with ethyl acetate – formic acid – glacial acetic acid – water 15:1:1:2, detection by spraying with 10 % sulfuric acid in ethanol and heating at 105 °C for 5 min, evaluation under white light and at UV 366 nm. Identification of Radix Codonopsis by HPLC.

Lipophilicity parameters obtained by normal phase chromatography were validated in relation to corresponding reverse phase chromatographic data.
