The method was linear within the range of 1.4-3.7 µg/mL. Mean recovery values were 99.46-100.29 %. The method was validated for accuracy, precision, and specificity.

      High Performance Thin Layer Chromatography method for therapeutic drug monitoring of anti-epileptic drugs in serum

      Indian Drugs 42 (10), 665-670 (2005). HPTLC of carbamazepine, phenytoin, and phenobarbitone extracted with ethyl acetate from human serum, on silica gel with toluene - acetone 5:2. Quantification by absorbance measurement at 217 nm. Rf values were 0.20 for carbamazepine, 0.41 for phenytoin, and 0.49 for phenobarbitone. The linearity (r=0.998) was in the range of 100-2000 ng. LOQ was found to be 30 ng/spot for carbamazepine and 80 ng/spot for phenytoin and phenobarbitone. The accuracy was in the range of 88.5 to 98.1 % and the CV in range of 1.1 to 3.9 %. Intra day and inter day reproducibility was comparable and within the stated limits.

The method is developed for determination of the cough suppressant dextromethorphan hydrobromide in multi-component flu-relief solid caplets and liquid gel caps, and of the antihistamine clemastine fumarate in tablets. HPTLC on silica gel (pre-washed with dichloromethane – methanol 1:1) with ethyl acetate – methanol – ammonia 17:1:2 for dextromethorphan hydrobromide or dichloromethane – methanol – ammonia 90:10:1 for clemastine fumarate. Densitometry at 225 nm for dextromethorphan hydrobromide and at 216 nm for clemastine fumarate. Multiple samples of the three dosage forms were analyzed to confirm agreement between content of the active ingredients and label declarations.

      Alkaloid Composition of Chelidonium majus L

TLC of isoquinoline alkaloids (chelidonine, chelerythrine, sanguinarine, coptisine and berberine) in Chelidonium plant organs on silica gel with chloroform - methanol 2:1, and methylene chloride - methanol 97:3. Quantification by densitometry at UV 254 nm. Detection is very sensitive because of fluorescence of alkaloids without purification. Comparison with HPLC, showing that the TLC methodis the most simple, accurate, reproducible and convenient analytical technique for fast investigations and routine determination of Chelidonium alkaloids.

Quantitative determination by absorbance measurement at 295 nm. The method was found to be linear in the concentrate range of 1-5 ng/spot with recovery of 99.5-102.5 % for both compounds. The method was validated for linearity, accuracy, precision, repeatability, and specificity.
