Detection under UV 254 and 366 nm. Quantification by densitometry at 260 nm (absorbance).

      Separation of the enantiomers of dansyl-DL-amino acids by normal-phase TLC on plates impregnated with a macrocyclic antibiotic

      J. Planar Chromatogr. 13, 33-36 (2000). Direct racemic resolution of dansyl-DL-amino acids on silica gel plates impregnated with vancomycin (0.34 mM) as chiral selector. The mobile phase enabling successful resolution of most of the racemic dansyl amino acids were acetonitrile - 0.5 M aqu. NaCl 5:2 and 14:3. Detection with a fixed dual-wavelength UV light at 254 nm. Detection limit 2.1µg. New sorbent for separation of chiral compounds resulting in a simple, rapid, sensitive, cost-effective method.

Densitometry at 295 nm. The enantiomers of a-methylbenzoin and anisoin could not be separated.

      Direct enantiomeric TLC resolution of dl-penicillamine using (R)-mandelic acid and L-tartaric acid as chiral impregnating reagents and as chiral mobile phase additive

TLC of dl-penicillamine on silica gel 1) impregnated with L-tartaric acid; 2) impregnated with (R)-mandelic acid; and 3) with no impregnation but with chiral modifier added to the mobile phase. The mobile phases used were 1) acetonitrile - methanol - water 5:1:1; 3) ethyl acetate - methanol - water 3:1:1; and 2) acetonitrile - methanol - 0.5 % aqueous L-tartaric acid (pH 5) - glacial acetic acid 70:10:11:7. Detection by exposure to iodine vapors. The limit of detection of both was 120 ng/zone by use of L-tartaric acid (either by impregnation or added to mobile phase), and 110 ng/zone by use of (R)-mandelic acid. (R)-mandelic acid was found to be successful as a chiral impregnating reagent.

TLC of DL-amino acids and their L- or D-forms on home made silica gel plates impregnated with recrystallized vancomycin with n-butanol - methanol - water 25:6:5 after chamber saturation for 15 min. Detection by spraying with ninhydrin solution (0.2 % in ethanol). The vancomycin allowed for enantiomeric resolution of D- and L-amino acids.
