Quantification by densitometry at 600 nm, and using b-(4-hydroxy-3-phenyl), acetic acid as internal standard. The sample was cleaned up through a cyano minicolumn and extracted with ether.

Description of the powerful off-line combination of rapid, high-resolution overpressured-layer chromatography (OPLC) with the high sensitivity and rapidity of digital autoradiography (DAR), by which qualitative and quantitative analysis of radiolabeled metabolites in biological samples can be performed relatively quickly. OPLC/HPTLC of deramciclane (1R,2S,4R-(-)-N,N-dimethyl-2-[(1,7,7-trimethyl-2- phenylbicyclo[2,2,1]hept-2-yl)oxy]-ethaneamine 2-(E)-butanedioate) on silica gel with n-butanol - acetic acid - water 4:1:1, ethyl acetate - ethanol - triethylamine 25:25:2, acetonitrile - n-butanol - acetic acid - water 25:17:4:4 or 44:30:7:19. For comparison to the radioactivity signal, UV and fluorescence signals were evaluated at 254 and 366 nm.

      Quantitative determination of fluoxetine in human serum by high-performance thin-layer chromatography

      J. Sep. Sci. 33, 2206-2210 (2010). HPTLC of fluoxetine on silica gel with toluene - glacial acetic acid 4:5. Derivatization of fluoxetine with 4-dimethylamino-azobenzene-4-sulfonyl chloride. Quantitative determination by absorbance measurement at 272 nm. The hRF of fluoxetine was 45. Linearity was between 0.05-0.35 ng/µL. Detection and quantification limits were 230 and 700 ng/µL, respectively. The intra-day and inter-day precisions showed a %RSD lower than 3.9 %. Recovery (by standard addition) was 94.7 %. The method was designed to cover the usual serum concentration level of patients taking the drug.

Elution of the indicator with chloroform - methanol 8:2 and of cholesterol glucuronide and ganglioside with chloroform - methanol 5:5.
