For quality control, TLC on silica gel 1) for Gastrodia elata Blume and the standard gastrodine, with chloroform – ethyl acetate – methanol – formic acid 80:10:30:1, detection by spraying with 10 % phosphomolybdic acid in ethanol and heating at 110 °C until the zones are visible in daylight; 2) for Epimedium davidii Franch and the standard icariin, with benzene – methanol – glacial acetic acid 45:8:4, detection by spraying with 10 % aluminium chloride in ethanol and letting stand for 40 min at room temperature, evaluation under UV 366 nm; 3) for Salvia miltiorrhiza and the standard protocatechuic aldehyde, with chloroform – acetone – formic acid 25:10:4, detection by exposure to ammonia vapors for 15 min and viewing under UV 254 nm. For quality control and to prevent counterfeiting TLC of herbal extracts and the standard euparin on silica gel with n-hexane – chloroform – methanol 20:10:1, detection by spraying with 1 % aluminium chloride in ethanol and heating until the zones are visible in daylight, and under UV 366 nm.

      Spectrodensitometric determination of some pharmaceutical mixtures containing proton pump inhibitors

HPTLC of omeprazole (1), panoprazole (2), rabeprazole (3), and lansoprazole (4) in binary mixtures with domperidone (5) (method A) and ternary mixture containing omeprazole (1), tinidazole (6), and clarithromycin (7) (method B) on silica gel with ethyl acetate - methanol - benzene - acetonitrile 5:4:8:3 for (A) and chloroform - n-hexane - methanol 3:4:1 for (B). Quantitative determination by absorbance measurement at 293 nm for (1)/(5) and (3)/(5), 286 nm for (4)/(5), 287 nm for (2)/(5), and 310 nm for (1)/(6)/(7). The hRF values for (1) to (5) in (A) were 54, 55, 55, 60 and 33, respectively, and for (1), (6) and (7) in (B) were 68, 22 and 48, respectively. Linearities for (1) to (7) were in the range of 240-1440 ng/zone, 180-1080 ng/zone, 240-1440 ng/zone, 240-1440 ng/zone, 120-720 ng/zone, 30-180 ng/zone and 2250-6000 ng/zone, respectively. The intermediate/interday/intra-day precisions were below 4 % (n=6). Recoveries ranged from 98.1 to 103.1 % for (1) to (7).

Zhiqiao Jianwei Keli granule is a herbal TCM for treatment of stagnated heat of liver and stomach, stomach pain and stomach cramps etc. For quality control, TLC on silica gel 1) for Fructus aurantii, with the lower phase of chloroform – methanol – water 13:6:2, detection by spraying with 3 % aluminum trichloride in ethanol and heating at 105 °C for 5 min and evaluation under UV 366 nm; 2) for Corydalis yanhusuo W. T. Wang ex Z. Y. Su et C. Y. Wu, with toluene – acetone 9:2, detection by exposure to iodine vapors for 3 min and evaluation under UV 366 nm; 3) for Forsythia suspensa (Thunb.) Vahl, with chloroform – methanol – glacial acetic acid 17:2:1, detection by spraying with 5 % vanillin in sulfuric acid – ethanol 1:4 and heating at 105 °C until the zones are visible in daylight. For determination of tetrahydropalmatine, TLC on silica gel with toluene – acetone 9:2 with chamber saturation for 30 min, detection by exposure to iodine vapors for 3 min. Evaluation under UV 366 nm and quantification by densitometric measurement at 343 nm using external standard calibration. Validation by investigating the linearity range of the calibration curve (55-330 ng/zone, r=0.996, n=6), the stability within 6 hours (%RSD 1.9 %, n=7), the precision (%RSD 1.3 %, n=6), and the repeatability (%RSD 0.8 %, n=6). Recovery was 98.0 % (%RSD 1.9 %, n=5).

      Extraction of artemisinin, an active antimalarial phytopharmaceutical from dried leaves of Artemisia annua L

      Chromatogr. Res. Int. 10, 1-11 (2014). HPTLC of artemisinin of Indian Artemisia annua L. was performed on HPTLC foil silica gel with n-hexane - ethyl acetate 3:1 in a twin-trough chamber up to a migration distance of 85 mm. Detection with anisaldehyde sulphuric acid reagent followed by heating at 110 °C for 10–15 min. Quantitative determination of artemisinin by densitometry at 540 nm. The hRf value of artemisinin (pink colored) was 28. The linearity was between 400 to 2800 ng/spot with a correlation coefficient of 0.9975. The precision (%RSD) was ≤3.3% (n=3). The LOD and LOQ were 40 ng/zone and 80 ng/zone, respectively. Recoveries were between 74 and 105 % for 0.6 mg/mL. Artemisinin content was highest in the leaf extract; no artemisinin was detected in the root extract.

For quality control, TLC on silica gel (1) for Cyperus rotundus, with petroleum ether (60-90 °C) – ethyl acetate 15:1, detection at UV 254 nm; (2) for Atractylodes macrocephala Koidz. and atractylone with petroleum ether (60-90 °C) – ethyl acetate 50:1, detection by spraying with 5 % vanillin in sulfuric acid – ethanol 1:4, followed by heating until zones are visible; (3) for Angelica dahurica (Fisch. ex Hoffm.) Benth. et Hook. f. ex Franch. et Sav. and imperatorin with petroleum ether (60-90 °C) – diethyl ether 1:1, detection at UV 366 nm; (4) for Epimedium brevicornum Maxim and icariin with ethyl acetate – butanone – formic acid – water 10:1:1:1, detection at UV 366 nm before and after spraying with 3 % aluminium trichloride in ethanol.
