      Planar-chromatographic fingerprint of German propolis

      CBS 111, 13-15 (2013). HPTLC of propolis on silica gel in a twin trough chamber with 8 mL of n-hexane – ethyl acetate – glacial acetic acid 5:3:1 and preconditioning with 5 mL hydrochloric acid (37 %) in the opposite trough using filter paper wetted with the hydrochloric acid shortly before chromatography. Migration distance was 65 mm. Detection under UV 366 nm after dipping in 0.5 % methanolic 2-aminoethyldiphenylborane reagent followed by drying and dipping in 5 % methanolic polyethylene glycol solution. Different types of German propolis were found, an orange, a blue and mixed types. Comparison with HPTLC fingerprints of the possible origin plants showed that the orange type most likely derives from Populus nigra. The blue type correlated to a certain extent with Populus tremula as well as both types with Aesculus hippocastanum L. Confirmed by MS the following substances were found in the orange type: pinocembrin, galangin, caffeic acid phenethylester and chrysin. The blue type contained p-cumaric acid and ellagic acid. Caffeic acid, quercetin and apigenin appeared in both types.

Zaodi Anshen Wan pill is herbal TCM for treatment of neurasthenia, fatigue syndrome, depression, insomnia, dizziness and tinnitus, palpitation, etc. For quality control, HPTLC on silica gel 1) for Ziziphus jujuba Mill. var. spinosa, with water-saturated n-butanol, detection by spraying with 1 % vanillin in sulfuric acid – ethanol 1:4, evaluation under UV 366 nm; 2) for Radix polygoni multiflori preparata and the standards emodin and physcion, with petroleum ether (30-60 ˚C) – ethyl formate – formic acid 15:5:1, detection by exposure to ammonia vapors, evaluation under UV 366 nm. Analysis of Schisandra chinensis, schisandrin, deoxyschizandrin and gamma-schizandrin B by HPLC.

      Development and validation of a high-performance thin-layer chromatographic–densitometric method for the quantification of swertiamarin in traditional bitters and formulations

      J. Planar Chromatogr. 28, 61-66 (2015). HPTLC of swertiamarin in extracts of Enicostemma littorale and Swertia chirata on silica gel with ethyl acetate – methanol – water 16:3:1. Quantitative determination by absorbance measurement at 246 nm. The hRF value of swertiamarin was 58. Linearity was between 200 and 1200 ng/zone. The intermediate intra-day and inter-day precisions were below 2 % (n=6). The LOD and LOQ were 38 and 115 ng/zone, respectively. Recoveries were in the range of 98-100 %.

For quality control, TLC on silica gel 1) for Angelica sinensis and Ligusticum chuanxiong Hort, with n-hexane – ethyl acetate 19:1, detection at UV 366 nm; 2) for Atractylodes lancea (Thunb.) DC., with petroleum ether (60-90 °C) – ethyl acetate 19:1, detection by spraying with 5 % p-dimethylaminobenzaldehyde in sulfuric acid – ethanol 1:4 and evaluation under white light; 3) for Corydalis yanhusuo W. T. Wang ex Z. Y. Su et C. Y. Wu and the standard tetrahydropalmatine, with n-hexane – chloroform – methanol – triethylamine 10:6:1:1, detection by exposure to iodine vapors until the zones are clearly visualized, evaluation under white and at UV 366 nm; (4) for Pericarpium Citri reticulatae and the standard hesperidin, with chloroform – acetone – methanol 5:1:1, detection by spraying with 3 % aluminium trichloride in ethanol and heating mildly until the zones are visible under white and at UV 366 nm. Quantification of hesperidin by HPLC.

For quality control, TLC on silica gel 1) for Equisetum hyemale L. and the standard kaempferide with cyclohexane – ethyl acetate – formic acid 20:10:1, detection by spraying with 5 % aluminum trichloride in ethanol and evaluation at UV 366 nm; 2) for Heartleaf Houttuynia Herb with benzene – chloroform – acetone 8:5:7, detection at UV 366 nm. Quantification of kaempferide by HPLC.

      Application of analytical validated high-performance thin-layer chromatographic technique for the multicomponent analysis of cardiovascular drug combos in pharmaceutical dosage form

TLC of rosuvastatin (1) and fenofibrate (2), rosuvastatin (1) and ezetimibe (3) and rosuvastatin (1) and aspirin (4) (in three different pharmaceutical combination products) on silica gel with toluene - acetone - acetic acid 30:10:1. Quantitative determination by absorbance measurement at 254 nm. The hRF values for (1) to (4) were 19, 83, 43 and 53, respectively. Linearity was in the range of 500-4000 ng/zone for all four drugs. LOD and LOQ were 77 and 234 ng/zone for (1), 38 and 116 ng/zone for (2), 108 and 327 ng/zone for (3) and 144 and 438 ng/zone for (4), respectively. The intermediate precision was below 1 % (n=3). Average recovery ranged between 98.9 and 99.4 % for (1) to (4).

      Effects of Inonotus hispidus extracts and compounds on human immunocompetent cells

The presence of hispidin (a styryl pyrone) and hispolon in a methanolic Soxhlet extract of powdered Inonotus hispidus was verified by TLC on silica gel with ethyl acetate – methanol - water 200:27:20. Detection under UV light before and after derivatization with Natural Product/PEG reagent, and with anisaldehyde sulfuric acid reagent (followed by heating for 5 min at 110 °C). The two phenolic compounds were confirmed and quantified by HPLC.
