      Stability-indicating chromatographic methods for the determination of sertindole

TLC of sertindole subjected to stress stability studies, including acid, alkali, oxidative, photolytic and thermal degradation, on silica gel with methanol – ethyl acetate – 33 % ammonia 10:90:1, detection under UV 254 nm (the hRf was 71 for sertindole and 16 for its oxidative degradation product), identification by IR and GC/MS, quantification by densitometry at 227 nm with a linearity range of the calibration curve from 2 to 14 µg/band. The intermediate precisions and repeatabilities were < 2%. The method is suitable for quality control laboratories as stability-indicating method and for routine analysis without any preliminary separation step.

      Ionic liquids as separation enhancers of haloperidol and its two metabolites in high-performance thin-layer

HPTLC of haloperidol (1) and its two metabolites: reduced haloperidol (2) and 4-(4-chlorophenyl)-4-hydroxypiperidine (3) in plasma samples on silica gel with acetonitrile – water – 1-ethyl-3-methylimidazolium tetrafluoroborate 100:100:3. Quantitative determination by absorbance measurement at 240 nm for (1) and 250 nm for (2) and (3). The hRf values for (1) to (3) were 34, 28 and 40, respectively. The identity of the analytes was additionally confirmed by elution of the zones with a TLC–MS interface connected to a triple quadrupole MS equipped with an electrospray ionization source (ESI). Linearity was in the range of 0.6-2.1 μg/zone for (1), 3.5-6.6 μg/zone for (2) and 2.4-5.4 μg/zone for (3). The intermediate precision was below 4.8 % (n=6). The LOD and LOQ were 180 and 548 ng/zone for (1), 316 and 957 ng/zone for (2) and 392 and 1189 ng/zone for (3), respectively. Recovery was in the range of 95.4-99.6 % (intra-day) and 96.1-103.2 % (inter-day) for all tested compounds.

Detection limit between 62.5-500 ng, depending on the impurity. The proposed procedure shows several advantages over tile related compounds test of the US Pharmacopoeia.
