Quantification by densitometry at 215 nm. Repeatability of measurement (peak area) was 0.89%; repeatability of sample application (peak area) was 2.89%.

      Thin-layer chromatography and mass spectrometry for screening of biological samples for drugs and metabolites

Description of a method for off-line coupling of TLC and electron-impact ionization mass spectrometry (EIMS) which is well suited for routine forensic and toxicological investigations of a large number of samples incl. discussion of advantages and drawbacks of this approach. Several TLC systems for 493 compounds of forensic and toxicological interest are described and eight-peak mass spectra from full EI mass spectra are listed. TLC on silica gel with 1) methanol - 25% NH3 200:3, 2) cyclohexane - toluene - diethylamine 15:3:2, 3) chloroform - methanol 9:1, 4) chloroform - acetone 8:2, 5) ethyl acetate - methanol - 25% NH3 17:2:1, and 6) ethyl acetate. Reagents used for color reactions were: Marquis, modified Dragendorff, Ludy Tenger, potassium iodoplatinate, palladium chloride, ferric chloride-sulfuric acid, mercury nitrate-mercury sulfate, diphenylcarbazone, fast blue B, Bratton-Marshall (Clarke's Isolation and Identification of Drugs, 2nd edn, The Pharmaceutical Press, London, UK, 1986). TLC plates for biological samples were pretreated with a solution of methanol and 1.5 aqueous ammonia and reactivated for 2 h at 150°C in a clean oven and subsequently cooled in a clean closed system.

      Densitometric and videodensitometric determination of nadolol and pindolol in tablets by quantitative HPTLC

HPTLC of nadolol (5-[3-[(1,1-dimethylethyl)amino]-2-hydroxypropoxy]-1,2,3,4-tetrahydro-2,3-naphthalenediol) and pindolol (1-(1H-indol-4-yloxy)-3-[(1-methylethyl)amino]-2-propanol) on silica gel with ethyl acetate - methanol - acetic acid 49:49:2. Detection and quantitation by densitometry at 270 nm and by video densitometry at 254 nm. In a densitometric procedure the linearity range was 0.2-1.2 µg/10 µL; in a videodensitometric assay the linearity ranges were 2.0-12.0 and 0.2-1.2 µg/10 µl for N and P, respectively. Simple, precise, and rapid procedure.

Densitometry at 270 nm. The proposed method is highly accurate, selective, and precise and can therefore be used for a routine quality-control analysis.
