      Analysis of decongestant and antihistamine pharmaceutical tablets and capsules by HPTLC with ultraviolet absorption densitometry

HPTLC of chlorpheniramine maleate, methscopolamine nitrate, pseudoephedrine hydrochloride, brompheniramine maleate, phenylpropanolamine hydrochloride, and guaifenesin on silica. PE, CM, and BM were developed to a distance of ca. 6 cm with 1-butanol - deionized water - acetic acid 7:2:1; plates containing PP were developed to a distance of ca. 6 cm with ethyl acetate - methanol - conc. NH3 17:2:1. All developments were performed at 20°C and 60% relative humidity with chamber saturation. Quantification by densitometry at 277 nm for PE, at 264 nm for CM, at 265 nm for BM, and at 209 nm for PP. It has been demonstrated that the new HPTLC method achieved recoveries as a percentage of tablet label values, standard deviations for replicate analyses and recoveries of spiked samples that compare very favorably with data reported on the HPLC analyses of drugs. Advantages of the method: Internal standards are not required, high sample throughput, very low solvent usage per sample, accuracy and precision are more than satisfactory for performing pharmaceutical analysis with validation according to GLP and GMP recommendations.

Densitometry in absorbance/reflectance mode at 240 nm for sulpiride and impurity B; after spraying with ninhydrin solution, heating at 80°C for 10 min scanning of impurity A was done at 240 nm and 500 nm. Simple, accurate, reproducible and quick TLC procedure.
