One-dimensional TLC of 8 quinolizidine alkaloids (sparteine, a-isosparteine, retamine, hydroxylupanine, cytisine, methylcytisine, sophocarpine, lupanine) on silica gel with hexane - diethyl amine 100:0 to 1:1, on silica gel and DIOL with chloroform - methanol - NH3 100:0:1, 95:5:1 to 85:15:1, and on DIOL and RP-18 with acetonitrile - water - hydrochloric acid 30:100:5, 30:100:10, 30:100:15 and 30:100:20.. 2D-TLC on DIOL with chloroform - methanol - NH3 85:15:1 (I) in the first direction and after drying at room temperature with acetonitrile - water - hydrochloric acid 30:100:7 (II) in the second direction. In adsorbent TLC - diol plates were used in the first direction with mobile phase I. Then the alkaloids were transferred by a specially designed device on the second RP-18 plate with the mobile phase II. The same phase was then used for the second development in the perpendicular direction. Visualization by spraying with Dragendorff reagent and documentation by photography.

      HPTLC investigations on St John’s Wort

HPTLC of flavonoids, hypericin and pseudohypericin on silica gel and diol phases (for quantification of hypericin) respectively, with ethyl acetate - dichloromethane - formic acid - acetic acid - water 100:25:10:10:11. Detection by dipping the warm plate in modified natural products reagent (0.5 % in ethyl acetate). Selective HPTLC of hyperforin on silica gel with toluene - dichloromethane 4:1. Visual detection by spraying with Godin reagent. Quantitative determination by absorbance measurement at 310 nm without derivatization.

      Densitometric standardization of herbal medical products containing Evolvulus alsinoides by quantification of a marker compound

      J. Planar Chromatogr. 18, 234-239 (2005). TLC of Evolvulus alsinoides extracts and the marker EA 1, i.e. 3beta,23,24-trihydroxyolean-12-en-28-oic acid, on silica gel with n-hexane - ethyl acetate 7:3 in a saturated chamber. The marker EA 1 was detected under UV light at 366 nm as a yellow fluorescent band of Rf 0.8. Quantitative determination by absorbance measurement at 232 nm (=UV max of EA 1). Limit of detection 11.6 ng; satisfactory recovery from 93.3 to 96.6 %. The marker was isolated from the aerial parts of Evolvulus alsinoides by preparative TLC.

Detection under UV light at 254 nm and by spraying with modified anisaldehyde - sulfuric acid spray reagent. For selective detection of cannabinoids, the plate was sprayed with 0.5 % fast blue B salt in water, followed by 0.1 M NaOH.
