Yijingling oral liquid is a herbal TCM preparation for mind and body tranquilization and nourishing the liver and kidney and is prescribed clinically to treat insomnia, amnesia, and the climacteric syndrome. For quality control, TLC on silica gel 1) for Radix Polygoni Multiflori with toluene – ethyl acetate – formic acid 20:2:1, detection by exposing to iodine vapor and viewing in daylight; 2) for Fructus Psoraleae with n-hexane – ethyl acetate 3:1, detection by spraying with 10 % KOH in ethanol and viewing under UV 366 nm.

      New salting out stability-indicating and kinetic thin-layer chromatographic method for determination of glimepiride and metformin HCl binary mixture

TLC of two antidiabetic drugs, glimepiride and metformin HCl in pure and tablet form as a binary mixture, or subjected to acidic, basic, oxidative and photo-degradative stress, by using an optimized salting out stability-indicating and kinetic TLC technique (SOTLC) on silica gel with aqueous ammonium sulfate – acetonitrile 7:3. The hRf value of glimepiride was 26±2 and of metformin HCl 73±2. Quantitative determination by densitometry at 237 nm in the concentration range of 60-1400 ng/zone for both drugs. The correlation coefficient of the calibration curve was 0.996 for glimepiride and 0.997 for metformin HCl.

      All on one high-performance thin-layer chromatography plate

      J. Chromatogr. A, 1572, 145-151 (2018). Introduction of on-surface reactions as a new strategy for rapid structure elucidation. This was illustrated by a miniaturized on-surface synthesis-guided identification of two new degradation products (impurities) occurring in a pharmaceutical formulation of the anti-cancer drug ifosfamide, especially in the presence of urea. The respective reagents were applied in the nanomole scale accurately and automated on a HPTLC silica gel plate. After a fast reaction in the start zone, the plate was developed, followed by online elution to high-resolution MS, whereby the on-surface reaction highlighted the impurities. As proof of concept and for benchmarking, it was compared to a reaction mixture obtained from conventional preparative synthesis in a round-bottom flask as well as to different formulations. Image evaluation was performed by videodensitometry. Discussion of the advantages such as: 1) the combination of all relevant steps on one HPTLC plate and its resulting efficiency made surface synthesis on chromatographic phases an optimal tool for signal highlighting in MS, and thus for the assignment of impurities in drugs; 2) the miniaturization of the chemistry process scale down to the μg-level per synthesis (in total 30-60 μg chemicals/reaction), setting a new state-of-the-art standard; 3) the contribution to a greener chemistry by reducing the consumption of chemicals and enhancing the analytical efficiency, when adapted for the quality control of any other chemical product.

      Determination of methyl nicotimate in pharmaceutical creams by high-performance thin-layer chromatography

Detection by spraying with 0.1 % solution of the ammonium salt of 8-anilino-naphthalene-1-sulfonic acid in water. Quantification by densitometry at 263 nm. 1) 3-pyridine carboxaldehyde was used as internal standard. 2) After quantification of methyl nicotinate the same plate was used for the identification of the cream base excipients with methanol - chloroform 3:2.
