      Optimization of separation of (+)-catechin and (-)-epicatechin on cellulose TLC plates

Improvement of separation TLC of (+)-catechin and (-)-epicatechin on cellulose with 1-butanol - water - acetic acid 4:2:1, developed in horizontal chambers (tank and sandwich configuration), with and without preconditioning. Prewashing of the plates, preconditioning, and the type of developing chamber had a large effect on the chromatographic results. Without prewashing of the TLC plate unsatisfactory or misleading results were obtained. Anisaldehyde-sulfuric acid dipping reagent proved to be more selective than vanillin - phosphoric acid as detection reagent for flavan-3-ols; the latter was, however, more sensitive.

Alternatively, a solution of 3 g bismuth nitrate in 100 mL acetone can be used; in addition observation under UV 254 and 366 nm. Densitometry at 417 nm for allicin and at 392 nm for disulfides.

Quantitative determination by absorbance measurement at 310 nm. Calibration was linear in the range of 10-100 µg/mL. The method was repeatable and precise with RSD between 0.98 and 1.59 % intra-day and between 3.41 and 5.56 % inter-day. Limits of detection and quantification were 3 and 6 µg/mL.
