For quality control, TLC on silica gel 1) for Astragalus membranaceus (Fisch.) Bunge., with n-butanol – ethyl acetate – 10 % diluted ammonia 4:1:5, detection by spraying with 10 % sulfuric acid in ethanol and heating at 105 °C and viewing in daylight; 2) for Pericarpium Citri Reticulatae, with toluene – ethyl acetate 3:2, detection by heating at 105 °C for 3 min and viewing under UV 366 nm; 3) for Paeonia veitchii, with toluene – ethyl acetate – isopropanol – methanol – ammonia 20:10:5:5:1, detection by exposure to ammonia vapors for 15 min and spraying with 5 % vanillin in sulfuric acid – ethanol 1:4 and heating at 105 °C, viewing in daylight. Quantification of astragaloside A by HPLC.

Gangmei Ganmaoling keli granule is herbal TCM for the treatment of colds, upper respiratory tract infections etc. For quality control TLC on silica gel 1) for Holly roots, with toluene – ethyl acetate – glacial acetic acid 10:3:1, detection under UV 366 nm and under daylight after exposing to ammonia vapors; 2) for Dendranthema indicum and the standard buddleoside, with ethyl acetate – butanone – chloroform – formic acid – water 15:15:6:4:1, detection by spraying with 2 % aluminum trichloride in ethanol and heating until the zones are visible under UV 365 nm; 3) for Isatis tinctoria and the standard arginine, with n-butanol – glacial acetic acid – water 19:5:5, detection by spraying with 2 % ninhydrin in acetone and heating at 105 ˚C until the zones are visible in daylight.

Ganoderma lucidum is a class of medicinal fungi considered by TCM physicians as the “god medicine” effective for inhibiting tumors, reducing free radicals, slowing down aging, improving the immunity, and having anti inflammation, hypolipidemic and hypoglycemic effects. Ganoderma lucidum polysaccharide is one of the most effective compounds of Ganoderma lucidum. Compared to GC and HPLC, this TLC method has the advantage of simpler sample preparation and multiple sample analysis which makes it more feasible. TLC of the hydrolysate of the sample and of standards glucose, L-rhamnose monohydrate, galactose, mannose and glucuronic acid on microcrystalline cellulose phase, with n-butanol – ethyl acetate – pyridine – water 6:1:5:4 (optimized from 6 different mobile phase systems), first run over 2 cm and second run over 8 cm. Detection by spraying with a solution of o-phthalic acid – n-butanol saturated with water – aniline 16:1000:9 and heating at 110 °C for 5 min and viewing in daylight.

Quantification of berberine hydrochloride by HPLC.

Redouté with chloroform – methanol – ammonia 40:10:1, detection at UV 254 nm; (3) for Terminalia chebula Retz. with chloroform – ethyl acetate – formic acid 6:4:1, detection by spraying with 2 % ferric chloride in ethanol, evaluation under white light.

      Development and validation of a high-performance thin-layer chromatographic method for the determination of the biomarker ?-amyrin in the leaves of different Ficus species

HPTLC of beta-amyrin in the leaves of five different Ficus species (F. carica, F. nitida, F. ingens, F. palmata and F. vasta) on silica gel with toluene - methanol 9:1. Detection with p-anisaldehyde reagent followed by heating. Quantitative determination by absorbance measurement at 550 nm. Precisions (intra- and inter-day) were 1.6-1.8 % and 1.7-1.8 % respectively (n=6). The hRF value was 58. Linearity was between 100 and 900 ng/zone. The LOD and LOQ were 32 ng and 96 ng/zone, respectively. Recoveries were 97.6-98.3 %.

      A new TLC bioautographic assay for qualitative and quantitative estimation of lipase inhibitors

      Phytochem. Anal. 27, 5-12 (2015). TLC-bioautography (editors note: TLC-autography) of lipase inhibitors in the stems of Flammulina velutiper and Pleurotus eryngii and the roots of Panax quinquefolius on silica gel with chloroform - ethyl acetate - methanol - water 15:40:22:9. The plates were dipped in β-naphthyl myristate solution, followed by air-drying at room temperature and dipping into lipase solution (60 U/mL), followed by incubation at 37 °C for 20 min. After dipping in 1 mg/mL of Fast Blue B salt solution, the plates were incubated again. Detection at 606 nm. Relative lipase inhibitory activity was calculated as orlistat equivalents. LOD was 0.01 ng/zone for orlistat. The intermediate precision was below 4.9 % (n=3). Recovery was in the range of 92-105 %.
