      Use of database of plots of pesticide retention (Rf) against mobile-phase compositions for fractionation of a mixture of pesticides by micropreparative Thin-Layer Chromatography

Relationships between Rf values and mobile phase composition have been determined for urea herbicides and fungicides in normal-phase systems (NP) of the type silica-nonpolar or weakly polar diluent (heptane, toluene, diisopropyl ether) – polar modifier (ethyl acetate, tetrahydrofuran, dioxane, ethyl-methyl ketone and 2-propanol). These relationships constitute a retention database which has enabled to choose optimum systems for preliminary fractionation of a multicomponent mixture of pesticides by zonal micropreparative TLC. The mixture was applied from the edge of the layer in the “frontal + elution” mode which increased the separation efficiency because of displacement effects. The separated simpler fractions were applied to a silica plate and rechromatographed. The plate was videoscanned, furnishing a real picture of the plate showing preliminary separation of the simpler pesticide fractions. Complete separation of the fractions was carried out by two-dimensional thin-layer chromatography on plates with chemically bonded-cyanopropyl silica stationary phase using non-aqueous eluent in the first direction and aqueous reversed-phase eluent in the second direction.

      A new spray reagent for selective detection and quantification of dichlorvos in bluish tinged maize grains by TLC-spectrophotometry

      J. Planar Chromatogr. 24, 113-115 (2011). TLC of dichlorvos on silica gel with cyclohexane - acetone - methanol 16:6:1. Detection by spraying with 2 % sodium hydroxide solution and subsequent spraying with 2 % thiobarbituric solution followed by heating at 90 °C for 10 min. The hRf of dichlorvos was 50. The limit of detection was approximately 18 µg/zone. Spectrophometric analysis of dichlorvos was performed by measuring the absorbance of the sample solution. The linearity was in the range of 50-350 µg/mL dichlorvos. On alkaline hydrolysis dichlorvos forms dimethylphosphoric acid and dichloroacetaldehyde; the latter reacts with 2-thiobarbituric acid to give a sharp pink spot. The reagent is selective for dichlorvos, and does not react with other organophosphorus, organochlorine, carbamate, and synthetic pyrethroid insecticides.
