Detection under UV 254 nm. Detection levels were 5 ng/zone each or 14-28 pmol. Desorption electrospray ionization mass spectrometry was investigated as a means to qualitatively identify and to quantify analytes directly from developed normal-phase TLC plates.

Quantitative determination by absorbance measurement at 360 nm. The hRF value for piperine was 90. The LODs and LOQs were 590 and 1800 ng/zone, respectively. Recoveries were between 98.1 and 99.2 %

      In vitro metabolism evaluation of the ergot alkaloid dihydroergotamine

      Planta Medica 82 (15), 1368-1373 (2016). The incubation mixture of dihydroergotamine with rat liver microsomes was dissolved in methanol – chloroform – ethyl acetate 1:1:1. Preparative TLC on silica gel with methanol – chloroform 1:9. Two zones (hRF 21 and 35) corresponding to produced metabolites were scratched off the plate, separated from the silica with methanol – ethyl acetate 1:1 and, after purification on cyclodextrin, identified through LC-MS-MS and proton-NMR as 5-hydroxyl-dihydroergotamine and 11-hydroxyl-dihydroergotamine, respectively.

      HPTLC quantification of cocoa ingredients and their changes during different chocolate manufacturing steps

      CBS 120, 9-11 (2018). HPTLC of oligomeric proanthocyanidins (PA), alkaloids, and anthocyanins in cocoa (fresh cocoa beans, roasted cocoa, cocoa mass and molded chocolate bars) on silica gel with ethyl formate – formic acid – water – toluene 60:8:6:3 to a migration distance of 70 mm. Detection by immersion in Fast Blue Salt B reagent (140 mg Fast Blue Salt B in 140 mL methanol, 10 mL water, and 50 mL dichloromethane), followed by drying. Evaluation under UV 254 nm and white light. Quantitative determination by absorbance measurement at 280 nm for alkaloids and 510 nm for anthocyanins and derivatized PA. Elution of target zones after derivatization with Fast Blue Salt B directly into the MS, mass spectra were recorded in positive ionization mode. In general, the HPTLC method provides good separation and quick quantification of the target substances. It was found that during fermentation and roasting the levels of the alkaloids and especially oligomeric PAs diminish.
