Optimized one-dimensional TLC on silica with chloroform - methanol - ethanol - acetic acid - water 48:8:4:12:1 has been employed for the determination of phospholipids in amniotic fluid. The developed plates were visualized by exposure to iodine vapor and the phospholipids of interest quantified in reflection/absorbance mode at 460 nm. Detection limits and recoveries for four phospholipids, the repeatability and the reproducibility of the method are reported. The method is sensitive and specific, and has been clinically applied in the determination of fetal lung maturity.
