      Planar chromatographic procedures in the quality control of Venticute(r) - a human homologous recombinant surfactant

Quantification by absorbance measurement at 365 nm. Intermediate precision 1-4%, recovery 98-103%, limit of quantification 0.1-0.4% (ca. 15-40 µg/spot). Immuno TLC on diol plates with chloroform - methanol - 8% NH3 65:30:6. Immunological staining according to conventional western plot. Visualization via enhanced chemiluminescence detection system. Densitometric evaluation via peak area of the x-ray film including background subtraction. Recovery 65-120% and 75-110% (inter-assay, n=6, 3 concentrations), repeatability 24-28% and intermediate precision 22-52% (n=6, 3 concentrations), detection limit 0,125 ng HCP (host cell derived proteins), quantitation limit 0,02% and specification limit 0,1%. This HPTLC procedure can also be used to separate the hydrophobic protein from impurities and modification (which can not be separated by HPLC). Visualization via coommassie brilliant blue (0,15 %). Repeatability ca. 10% (n=8).

TLC on silica gel with chloroform – methanol – water 40:10:1 for neutral and zwitterionic lipids and propanol – ammonia – water 15:1:1 for acidic lipids. The effect of tamoxifen on glycosphingolipid biosynthesis was evaluated by fluorescent precursor incorporation.

      Quality assessment of Mustadi Taila

Assessment of the quality of Mustadi Taila, an oil obtained from 8 plants (stems of Acacia chundra, Cedrus deodara, rhizome of Cyperus rotundus, seeds of Embelia ribes, roots of Glycyrrhiza glabra, Rubia cordifolia, Vitex negundo, Vetiveria zizanioides), and prescribed in Chakra Datta as a remedy for dental caries. TLC of the dichloromethane fraction of the aqueous oil extract with methanol – cyclohexane –dichloromethane 3:2:77. Evaluation at UV 254 and 366 nm revealed 10 prominent zones. Detection of alkaloids by spraying with Dragendorff’s reagent. Further properties of the oil (organoleptic as well as physico-chemical parameters, phytochemical screening, microbiological activity and heavy metals limits) were determined without TLC.

Detection with 20 % sulfuric acid in water - methanol 3:2, after drying the plate at 120 °C for 10 minutes, heating over metallic plate (200 °C, 10 minutes).
