      Thin-layer chromatography with chemiluminescent detection of enhancers of the luminol -H2O2-peroxidase system

Detection by spraying with a solution of 2mM luminol, 6 mM H2O2 and 0.744 U/mL peroxidase in a 0.094 M tris-HCl buffer (pH 8.5). Determination by densitometry with a single optical fibre that collected the emission signal to a spectrometer. Detection limit 38 ng and 14 ng respectively.

      A new visualization reagent for cation separation by TLC

TLC separation of Cr, Mn, Fe, Co, Ni, Cu, Zn, Ag, Cd, Pb on cellulose with methanol - benzene - ethyl methyl ketone - 1-butanol - 25% NH3 6:4:4:3:3, methanol - benzene - ethyl acetate - ethyl methyl ketone - 25% NH3 5:4:5:3:3, and methanol - benzene - ethyl methyl ketone - 1-butanol - 2 M HCl 6:4:4:3:3. Detection after drying by spraying with a new visualization reagent, a 0.01% solution of 9-formylacridine in dichloromethane. The plates were then heated at 120°C for 10 min and examined under UV 366 nm.

      Planar chromatography of heterocyclic thiols with detection by use of the iodine-azide reaction

      J. Planar Chromatogr. 19, 4-9 (2006). TLC and HPTLC of 2-thioguanidine and 6-mercaptopurine on silica gel with methanol in a horizontal DS-chamber. Spots were visualized with a freshly prepared solution of sodium azide and starch, adjusted to a pH within the range 5.5-6.0, and then exposed to iodine vapor. The thiols became visible as white spots on a violet-gray background. The iodine-azide reagent enabled detection of quantities in the range 1-80 pmol per spot.

      Coralyne cation, a fluorescent probe for general detection in planar chromatography

Fluorescence scanning densitometry of various analytes on HPTLC silica gel plates impregnated with a solution of coralyne cation, based on the increase or decrease, that the corresponding analyte induces on native coralyne emission at a given excitation wavelength. Compared to a procedure previously described for berberine cation, and Reichardt's dye probes, the sensitivity of coralyne in HPTLC detection of non-fluorescent, structurally different analytes (e.g. long-chain alkanes, alcohols, alkylbromides, neutral lipids) is superior.

      Modified ninhydrin reagents to detect colors of amino acid zones on thin-layer chromatographic plates

      J. Planar Chromatogr. 26, 517-520 (2013). TLC of amino acids on silica gel with 1-propanol - water 7:3. Detection by spraying with (1) 1 % 2-hydroxybenzenecarbaldehyde solution in propanone; or (2) 1 % 4-bromobenzenecarbaldehyde solution in propanone (2), followed by heating at 110 ºC for 10 min. After cooling, all plates (treated with (1) or (2)) were sprayed with ninhydrin (indane-1,2,3-trione monohydrate) reagent and air dried. Amino acid zones in various shades from yellow to brownish-red were detected (detailed table provided in paper). Plates were heated again at 110°C for 10 min and color changes were recorded. The LOD were determined visually by comparison of standard solutions (1 mg/L) and further dilution until no zone was seen. LOD for (1) and (2) were in the range of 0.1-1.0 µg/zone and 0.01-1.0 µg/zone, respectively.

Derivatization with 5 % perchloric acid in methanol.

Detection by spraying with a freshly prepared solution of formaldehyde (37%, 15 mL) + acetyl acetone (7.8 mL), diluted to 100 mL with acetate buffer (pH 4.7), and by heating at 100 °C for 10 min. Detection limits, 0.26 x 10-4 - 6.9 x 10-4 mmol. Comparison of the sensitivity of this reagent with ninhydrin reagent.
