      Visualizing agents for aromatic carboxylic acids in TLC

Description of the evaluation of 18 new visualizing agents for the detection of aromatic amines on a) silica, b) a mixture of silica and kieselguhr G with benzene - methanol - acetic acid 45:8:4, and c) polyamide with acetone - ethanol 4:1. For the separation on a) the best visualization results were obtained with bromocresol green, bromothymol blue, sodium hydroxide, thymol blue, and methyl red; for the separation on b) best results were obtained with thymolphthalein. The visualization effects on c) were not as good as on a) and b). The utility of any particular visualizing agent depends on the type of chromatographic support used. The composition and handling of the visualization reagents are given.

Fluorodensitometry at 360 nm. Improvement of the results by spraying the plates with a solution of 10% poly(ethylene glycol) 4000 in chloroform. Limit of detection in the lower picomole range.

      Application of iodine-azide reaction for detection of amino acids in thin-layer chromatography

      J. Chromatogr. A 1059 (1-2), 171-174 (2004). The iodine-azide reaction was employed to TLC detection of sulphur-containing derivatives of protein and some non-protein amino acids. The derivatization reaction with phenyl isothiocyanate (PITC) took place directly on the plate before the developing step. Subsequently, the plates were sprayed with a mixture of sodium azide and starch solution in NP-TLC and in the case of RP-TLC sodium azide solution with starch incorporated into mobile phase and then exposed to iodine vapour. The spots became visible as white spots on violet-grey background. The obtained detection limits of PTC-derivatives have been compared with other visualizing techniques commonly used in TLC practice (UV254 and iodine vapour). The iodine-azide system has been proved to be the most favourable and enabled to detect quantities per spot in the range of 1-60 pmol (HPTLC) and 3–100 pmol (TLC).

      A new way of using chemiluminescence in thin-layer chromatography

      J. Planar Chromatogr. 24, 357-359 (2011). TLC of benzo[a]pyrene on RP-18, RP-8, and RP-2 with acetone - ethyl acetate 7:3 in an unsaturated twin-trough chamber. Detection by dipping for 1 s into a solution of 250 mg bis(2,4-dinitrophenyl)oxalate in 40 mL of n-butyl acetate and 5 mL of hydrogen peroxide (upper phase). For videodensitometric evaluation the sheets were measured for 2 min and monitored over a period of 2 h. The reaction energy is transferred to the fluorescent compound (benzo[a]pyrene) which emits light while relaxing from its excited state. The advantage of this chemiluminescence reaction performed on a TLC plate is the improvement of the detection limit by extending the measuring time. The detection limit is 50 pg benzo[a]pyrene which is identical to that achievable in chemiluminescence HPLC. The calibration curve for benzo[a]pyrene was linear in the range of 50-8000 pg. The new procedure overcomes the problem of uneven illumination with CCD cameras.

      Specific detection of carbaryl on thin-layer chromatographic plates

A simple, sensitive and specific color reaction for the detection of carbaryl (detection limit 1µg) based on the condensation of ß-naphthol (product of carbaryl hydrolysis) followed by oxidation is described. Since other carbamate insecticides do not give the same red-violet color reaction it can be used routinely for specific detection in biological materials in forensic toxicology.

Visualization by spraying with fluorescamine solution and under UV 366 nm. Detection limit 100 ppb. Presentation also of the results from the industrial evaluation of the complete method.
