      TLC image analysis for determination of the piperine content of the traditional medicinal preparations of Bhutan

TLC of piperine, the bioactive constituent of black pepper (Piper nigrum), on silica gel with dichloromethane – ethyl acetate 9:1 at 30 °C in a twin-trough chamber saturated for 30 min. Detection under UV light at 254 nm and documentation with a digital camera. Based on the image a density profile plot was established by Scion Image software, which allowed to calculate the concentration of piperine by comparison of the peak areas of samples and piperine standards. The linearity was in the range of 24-84 ng/zone (r2=0.9927). The limits of detection and quantitation were 0.35 and 1.05 ng/zone, respectively. Precision (repeatability, n=6) and intermediate precision (2 days, n=12) both are below 2.6 %RSD. Recovery is between 96.7-101.4 %.

      Application of light scanning photoacoustic densitometer to thin-layer chromatography

Scanning with chopped light (128 Hz) in X and Y directions with quartz fiber optic over the quartz cell window (5 x1 0 cm). Detection of signal by a condenser microphone, position of zone by two potentiometers. A linear correlation between signal and amount of sample (methylene blue) was obtained from 4-20 mg. 8.0 mg gave 1.86 mV with S/N 5.23 on a layer (silica?) backed by aluminium foil, and 4.08 mV with S/N 11.7 on a silica glass plate.

      Thin-layer chromatography with supersonic jet fluorometric detection

Introduction of the 1st dye laser beam (275nm) from a small through-hole to desorb the chemical species on the sheet. The vaporized sample is entrained into a carrier gas of argon, and the gas is then expanded into a vacuum to form a supersonic jet. Detection of the sample molecules by fluorescence induced by the 2nd dye laser beam (366-375 nm) direct measurement of excitation spectrum and chromatogram for the sample spot. Detection limit 10 ng.

      Calibration in quantitative TLC based on weighed regression functions
