Presentation of the validation of a HPTLC procedure for quantification of caffeine with a Video Documentation System. Validation was performed according to the ICH guidelines, considering the special features of the method. The validation parameters were: selectivity, stability of the analyte, linearity, precision (repeatability and intermediate precision) and robustness. The obtained results were compared with those obtained by the slit-scanning densitometer. HPTLC of caffeine on silica with dichloromethane - methanol 9:1. Evaluation by densitometry at 270 nm. Repeatability 1.59% (densitometer) and 1.21% (CCD camera); intermediate precision 1.22% (densitometer) and 1.12% (CCD camera); robustness 1.47% (densitometer) and 1.37% (CCD camera).

Determination of the lipophilicity RM0 and log PTLC of thirteen novel, potentially biologically active, 12 H-quino[3,4- b ] [1,4] benzothiazinium salts by TLC on RP-18 with methanol – aqueous Tris buffer mixtures. RM values were linearly dependent on methanol concentration, and extrapolation of these to 0% methanol gave the lipophilicity RM0. log PTLC was obtained from RM0 by use of a calibration curve obtained for five standards of known experimental lipophilicity (log P). The lipophilicity log Pcalcd was calculated for the thirteen quinobenzothiazines by use of nine software products. The chromatographic lipophilicity RM0 can be used as a measure of the lipophilicity of the azaphenothiazine derivatives investigated.

Determination of parameters characterizing TLC systems. J.Liquid Chromatogr. 6, 81-93 (1983). Discussion of an equation, derived in terms of the thermodynamic formulation of TLC with mixed mobile phase. The parameters of this equation, describing solute-solvent interactions in thermolabile phase and phase-exchange equilibrium between molecules of both solvents are determined by using its linear forms. These linear relationships have been examined by applying the TLC data for eight different binary mobile phases.
